Historically, data have been transformed to facilitate plotting on linear plots such as Lineweaver-Burk (1/y versus 1/[S]), Hanes-Woolf ([S]/y versus [S]), or Eadie-Hofstee (v/[S] versus y). However, with the present availability of affordable nonlinear regression and graphing software packages such as GraphPad Prism,... [Pg.106]

Further drawbacks associated with the direct linear plot include the fact that this analysis does not readily lend itself to standard computerized graphing methods (for example, use of GraphPad Prism), although specialized software is available (Henderson, 1993). Of course, one of the major advantages of the direct linear plot is the ability to obtain kinetic constants by eye, without the need for a computer. However, for presentation purposes, the use of graphing software is still desirable. Furthermore, any behavior more complicated than simple, single substrate kinetics - for example, turnover in the presence of an inhibitor, or multisubstrate kinetics - caimot readily be shown on a direct linear plot. This is in contrast with the flexibility afforded by nonhnear regression approaches. [Pg.108]

Fig. 2. CD154 and IL-4 stimulation of CLL cells Inhibits dmg-Induced cell death. CLL cells were cultured on WT and CD154 NIH3T3 fibroblasts using either the continuous (a) or the discontinuous (b) protocol In the presence of bortezomlb (30 nM), PHA-767491 (10 pIVI), fludarablne (10 p.M), or vehicle control for 24 h. Cell death was analyzed using a BD FACSCanto I flow cytometer (BD Biosciences) using annexin V/Pi staining. Dataware analyzed using FlowJo ( Tree Star) and GraphPad Prism software. Fig. 2. All data were presented as mean SEM. The data were evaluated statistically for differences by one-way analysis of variance (ANOVA) followed by post hoc Tukey test for multiple comparisons, p < 0.05 was regarded as significant. Calculations were done using GraphPad Prism. [Pg.226]

The changes of observed parameters were evaluated using repeated-measures ANOVA with Bonferronis corrected J>ost hoc r-test for multiple comparisons of dependent variables. Pearsons correlation coefficient and test were used for the evaluation of dependence of quantitative variables, -values less than 0.05 were considered significant. All statistic analyses were performed using the software package GraphPad Prism 4.0. [Pg.359]

Add 50 pL/weU of H2SO4 2 M to stop the reaction. Read immediately the absorbance at 450 nm in an appropriate device. Data should be analyzed using GraphPad Prism nonhnear regression curve fitting or an equivalent software. [Pg.77]

All interactive mixtures were measured in triplicate and results were given as mean of three standard deviation. A paired t-test was performed for the results gained by Monckedieck et al. [11] to check for significance between the FPFs of the powder blends (GraphPad Prism, Version 6, Graph Pad Software Inc., La JoUa, USA). [Pg.533]

All statistical computations were performed using GraphPad Prism version 4.0a for Mac OS X (GraphPad Software, San Diego, California, USA). Values of experimental groups are shown as mean SEM unless otherwise stated. One-way ANOVA with Tukey s post-test analysis was used to determine statistical significance. Where appropriate, either two-tailed t-tests or Mann Whitney U tests were performed. A probability of P < 0.05 was considered to be statistically significant. [Pg.22]
