Liposomes, glycoprotein-incorporated

Studies of Con A induced agglutination of live cells are susceptible to complications such as irreversibility of the agglutinated cells upon aging (17), endocytosis of bound lectin (18) and effects of experimental conditions such as temperature, pH and ionic strength on membrane properties (J). Alternatively, the use of aldehyde-fixed cells (19), heatkilled cells (20), enucleated cells (21) and glycoprotein-incorporated liposomes (22) may lead to less problems being encountered.

Liposomes, in particular, may offer the future possibility of delivering enzymes (and other drugs) to specific body tissues. This could be attained by incorporating target molecules (e.g. specific receptors, glycoproteins or antibodies) in the liposome membrane, which selectively...

Naturally, liposomes as such are not unique in this context. Instead, the same approach can be used for other PEO-modified colloidal drug carriers, e.g. copolymer micelles. For example, Kabanov et al. have demonstrated specific targeting of fluorescein isothiocyanate solubilized in PEO-PPO-PEO block copolymer micelles conjugated with antibodies to the antigen of brain glial cells (a2-glycoprotein) (83, 84). Furthermore, incorporation of haloperidol into such micelles was found to result in a drastically improved therapeutic effect in mice, as inferred from horizontal mobility and grooming frequency studies.
