Glutaraldehyde stock

Paraformaldehyde, 0.2% glutaraldehyde m PBT (see above). Make this solution from a frozen, E. M. grade 25% glutaraldehyde stock (Sigma Grade 1) and freshly prepared paraformaldehyde. 

Preparation of 2.5% Glutaraldehyde Solution Prepare 10ml 2.5% solution of glutaraldehyde in 1 x SSC buffer by mixing 500 pi of glutaraldehyde stock solution with 9.5 ml of 1X SSC buffer. Always use freshly prepared solution. 

A stock of glutaraldehyde-activated BSA with a concentration of 10 mg/ml maybe stored frozen in aliquots. Activated BSA of high stability with about 20 moles aldehyde per mole BSA is made by a dialysis procediue given by Zegers et al. (1990) J Immimol Meth 130 195.

Glutaraldehyde solution Make a 2.5% solution of glutaraldehyde by diluting 25% stock solution 1/10 with PBS. This solution should be prepared fresh each time. 

Dissolve 10 mg of peroxidase in 0.2 ml of a freshly prepared 1 25 dilution of the stock glutaraldehyde solution in phosphate buffer and allow to stand at room temperature for 18 hr. 

Glutaraldehyde-Formaldehyde (see Note 16) 1. Dilute stock glutaraldehyde solution to 0.5% in PBS containing 2% formaldehyde (rrrNote 17). 2. Fix samples for 20 min to 1 h in fixative. 3. Rinse three times in PBS. 4. Process as usual. 

Retention-aid chemicals may be added, as well as defoamers (e.g. fatty-acid compounds), pitch-control agents, optical brighteners (usually stilbene derivatives) and, in some cases, fibre deflocculants. Different slimicides are also added to the stock for microbe control. A number of chemicals can be used for this purpose, such as 1,5-pentanediol, 2,2-dibromo-3-nit-rile propionamide, 2,2-dibromo-2-cyanoacetamide, 5-chloro-2-methyl-4-isothiazolin-3-one + 2-methyl-4-iso-thiazolin-3-one (Kathon CG) or glutaraldehyde. 

Glutaraldehyde solution 2% (v/v) glutaraldehyde. To make 1 liter of the solution, dilute 20 ml of anhydrous glutaraldehyde to final volume with deionized water. Prepare just prior to use. Store stock glutaraldehyde at 4°C. 

A 4% stock solution of lanthanum nitrate pH 7.6-7.8 was prepared as follows 4 g lanthanum nitrate was dissolved in 50 ml distilled water and the pH was adjusted to 7.6-7.8 with O.IM NaOH while stirring vigorously and slowly adding the base (Revel and Kamovsky 1967). The volume was is then brought to 100 ml with distilled water. One volume of this solution is added to 3 volumes of the fixative (e.g., 3.33% glutaraldehyde in 0.12 M sodium... 

Use EM-grade glutaraldehyde (available as a 50% stock in ampules Ted PeUa). Once opened, glutaraldehyde is good for 1 month if stored at 4°C.
