Gene Transfer Mechanisms

The most advanced method utilizes a binary vector system. One of the binary vectors is shown in Figure 7.11. This system depends on a small vector which contains the minimum elements required in cis. Other functions necessary for the gene transfer mechanism are donated from a separate helper, Ti plasmid. These features permit an easy introduction and maintenance of an exogenous DNA containing a manipulated gene.

Burch (1983) suggests that repair mechanisms cause a non neglectible complication for extrapolation from high to low doses and presents a modification of the linear-quadratic formula given above. CNTs with different characteristics, which will lead to differences in the mechanism of CNT metabolism, degradation or dissolution, clearance and bioaccumulation. On the other hand, most non-viral gene delivery systems today suffer from both limited levels of gene expression and an unfavourable toxicity profile due to their highly cationic surface character. Therefore, opportunities for CNT-based gene transfer systems are still ample. 

Ola Johnsborg is a research scientist. He received his Ph.D. in 2003 from the Norwegian University of Life Sciences. His Ph.D. work was on molecular mechanisms underlying regulation of bacteriocin production in Laaobacillus plantarum. Since then he has been studying horizontal gene transfer by means of competence for natural transformation in the human pathogenic bacterium Streptococcus pneumoniae.
