Gastric emptying cisapride effect

The effects of low-dose cisapride (0.1 mg/kg tds) on gastric emptjdng and the QT interval were studied in a donble-bUnd, placebo-controlled, crossover trial in 20 infants of low birth weights (29). Low-dose cisapride significantly improved gastric emptying without prolonging the QT interval.

Bethanechol is a synthetic derivative of acetylcholine and is not degraded by cholinesterase. Bethanechol has been used in humans to increase lower esophageal sphincter (LES) tone and to enhance gastric emptying. The cholinergic side-effects (salivation, abdominal discomfort) of bethanechol have reduced its use in human medicine since the introduction of metoclopramide and cisapride.

In dog stomach, benzamides and benzimidazolones are gastroprokinetic agents, that also promote gastric emptying [67, 82]. This effect is certainly mediated by acetylcholine release [82]. However, the effect of benzamides and in particular cisapride do not involve 5-HTj j -Rs [82]. Surprisingly, benzamides and 5-HT do not seem to mediate their effect via the same receptor. Alcohol Cisapride increases gastric emptying and can modestly increase serum alcohol levels. A modest 22% increase in the AUC of cisapride seen in one study. Unlikely to be significant, however the sedative effects might be accelerated. Unknown significance monitor patient for sedation. 1-5...
