Pearce and Lushnikova [78] used semipreparative HPLC method for the isolation of the three omeprazole metabolites produced by the fungi. Incubation of Cunninghamella elegans ATCC 9245 and omeprazole allowed putative fungal metabolite to be isolated in sufficient quantities for structural elucidation. The metabolites structures were identified by a combination of LC/MS and NMR spectrometric experiments. These isolates are used as reference standards in the confirmatory analysis of mammalian metabolites of omeprazole. In the LC/MS and LC/MS/MS analysis, components were separated by reversed-phase HPLC on a Hypersyl HyPurity (15 cm x 4.6 mm, 5 /im) column. The mobile phase consisted... [Pg.220]

The ASTM procedure describes in detail the conditioning of wood samples, dissolution of preservatives, impregnation, preparation of soil, bottles, sterilization procedures, inoculation of wood samples with one or more strains of wood-destroying fungi, incubation of samples and duration of the test, and so on. The principal readout of the test procedure is a loss of weight of the wood sample after its exposure to microbial cultures. [Pg.435]

The pathway of exposure (e.g., inhalation, lacerations) can also cause a significant change in the incubation time required as well as the clinical presentation of the disease. Diseases caused by fungi are not communicable and cannot be transferred directly from an infected individual to anyone else. [Pg.604]

After the six-month field test, the Prototype Unit was disassembled and the component parts were individually swabbed with sterilized cotton wools (4 cm2). Each samples were stored in 1 ml sterilized distilled water and 50 pi of samples were transferred to TSA and MEA plates. The TSA plates were incubated at 37 °C for 24 h and bacterial colonies were counted. The number of fungal colonies was determined from the MEA plates after incubating at 30 °C for 5 days. The results of the test are shown in Table 12.9-7. The results show that bacteria and fungi thrived near the air intake where they deposited on the grills, panel and around the intake slots. However once the microorganisms are drawn through the catalyst-coated desiccant wheel, the number of viable... [Pg.409]

In connection with our own work on the enzyme-catalysed hydrolysis of cyclohexene epoxide with various fungi we made the unexpected observation that the microorganism Corynesporia casssiicola DSM 62475 was able to interconvert the (1R,2R) and (1S,2S) enantiomers of the product, trans cyclohexan-1,2-dioI 25. As the reaction proceeded the (1R,2R) enantiomer was converted to the (1S,2S) enantiomer [20]. If the racemic trans diol 25 was incubated with the growing fungus over 5 days, optically pure (> 99 % e. e.) (1 S,2S) diol 25 could be isolated in 85% yield. Similarly biotransformation of cis (meso) cycIohexan-1,2-diol 26 yielded the (1S,2S) diol 25 in 41 % (unoptimized) yield (Scheme 11). [Pg.67]
