Fumaric acid esters psoriasis

In 83 patients with severe psoriasis vulgaris, the fumaric acid ester formulations Fumaderm initial and Fumaderm caused adverse effects in 62%, mainly flushing and gastrointestinal complaints, which decreased in frequency during the course of the study (6). [Pg.1453]

In 66 patients with severe psoriasis who took fumaric acid esters for up to 14 years, there were adverse events in 73%, usually mild and mainly consisting of flushing (55%), diarrhea (42%), nausea (14%), tiredness (14%), and stomach complaints (12%) (8). There was a relative lymphocytopenia in 76%, resulting in permanent withdrawal in four patients. There were transient eosinophUia in 14%, and moderate Uver enzyme rises were observed in 25%. [Pg.1453]

Tumorigenicity Fumaric acid esters have been used in the treatment of psoriasis since 1959, after Schweckendiek s description. They induce a shift from the T-helper 1 (Th-1) cytokine response to a Th-2 cytokine response and subsequent lymphopenia with low CD3 and CD4 counts.
