Fit securely to the lower end of the condenser (as a receiver) a Buchner flask, the side-tube carrying a piece of rubber tubing which falls well below the level of the bench. Steam-distil the ethereal mixture for about 30 minutes discard the distillate, which contains the ether, possibly a trace of unchanged ethyl benzoate, and also any biphenyl, CeHs CgHs, which has been formed. The residue in the flask contains the triphenyl carbinol, which solidifies when the liquid is cooled. Cartridges are inserted in series into plastic or stainless-steel tubular pressure housings of square cross section (Fig. 14). Feed flows parallel to the leaf surface. A permeate fitting secures each cartridge to the housing wall, which allows permeate egress and faciUtates sealing between concentrate, atmosphere, and permeate channels. 

With the shear-type coupling, the elastomeric element may be clamped or bonded in place, or fitted securely to the hubs. The compression-type couplings may be fitted with projecting pins, bolts, or lugs to connect the components. For proper operation of a centrifuge, only those tubes recommended by their manufacturer should be used. The material used for the tube must withstand the RCF to which the tube is likely to be subjected. Polypropylene tubes are generally capable of withstanding RCFs of up to 5000 Xg. The tubes should have a tapered bottom, particularly if a supernatant is to be removed, and should be of a size to fit securely into the rack to be centrifuged. The top of the tube should not protrude so far above the bucket that the swing into a horizontal position is impeded by the rotor.

Tips for dispensing in single-ehannel pipets have to be carefully considered. In cases in which small volumes (5 C20 pL) are pipeted, the pipet manufacturer s recommended tips should be used. It is essential that the tips fit securely on the pipets and that they can be pressed on firmly by hand (avoiding...

They must be robust enough to cope with sampling, transport, and storage conditions that are likely to be encountered. The lids of any container must fit tightly to prevent any loss of sample during transportation and storage if tipped and to prevent any entry of air especially if the sample is susceptible to oxidation. Alternatively the cap must fit securely to avoid sample loss.

You may need to obtain this flask from your instructor, along with a glass stopper or plastic cap that fits securely into the standard tapered joint. 

Pipettes The Pasteur pipette is shown in Figure 5.5A with a 2-mL rubber bulb attached. There are two sizes of pipettes a long one (9 inch) and a short one (5% inch). It is important that the pipette bulb fit securely. You should not use a medicine dropper bulb, because of its small capacity. A Pasteur pipette is an indispensable piece of equipment for the routine transfer of liquids. It is also used for separations (Technique 12). Pasteur pipettes may be packed with cotton for use in gravity filtration (Technique 8) or packed with an adsorbent for small-scale column chromatography (Technique 19). Although they are considered disposable, you should be able to clean them for reuse as long as the tip remains unchipped. 

Most mycotoxin TLC analyses are accomplished on silica gel plates, and Rf values vary, depending on the silica gel used. The plates most often used in FDA laboratories are precoated with a 0.2S-mm layer of silica gel 60 (E. Merck, Darmstadt). Plates from other manufacturers may work equally well. Some acidic mycotoxins, such as citrinin, are best identified on silica gel plates impregnated with oxalic acid. The plate is dipped in a 10% solution of oxalic acid in methanol for 2 min, and air-dried overnight in a hood. The fumonisins are analyzed by using reversed-phase C]g plates. The best resolution is obtained in an unlined, unequilibrated tank (11). A trough in the bottom of the tank is usually used to minimize the volume of solvent needed. The lid of the tank should fit securely. A leaky tank cover will result in nonuniform values across the plate. Development (10-12 cm) usually takes 60-90 min. 

Contents of the gas cylinder should be clearly identified. Color coding is NOT a reliable means of identification. Do not deface or remove any markings, tags, or stencil marks used for identification of contents attached by the vendor. Cylinders that do not bear a legibly written, stamped, or stenciled identification of the contents should not be used. They should be segregated and returned to the vendor as soon as possible. Caps used for valve protection should be kept on the cylinder except when the cylinder is in use. A cylinder s cap should be screwed all the way down on the cylinder s neck and should fit securely. The cap is for valve protection only. 

Assemble the safety clamp, ensuring that the upper clamp plate fits securely over the top end of the probe or coupon holder and prevents any upward movement of the same. 

The whole circuit (5x5cm) was attached on the top of a hard hat that can fit securely on the subject s head and provide a stable platform (Fig. 3). The hat was hght and comfortable enough to allow unencumbered and unrestricted movement of the neck.
