Rat F98 glioma cells were from R. Goodman, Ohio State University (Columbus, OH). The 9L gliosarcoma line was obtained from the Brain Tumor Research Center, University of California, San Francisco. Cells were maintained in 10% fetal calf serum in DMEM supplemented with penicillin/streptomycin and tested by the Gen-Probe Rapid Detection System (Fisher Scientific) to rule out mycoplasma contamination. Carboplatin coupling to a-cyclodextrin delays decomposition and increases bioavailabdity (155). Over 110 days, ethyl-cellulose microcapsules loaded with both agents at 2.2% (w/w) afforded 56% carboplatin release. Against the established rat intracranial F98 glioma, microcapsule delivery of a-cyclodextrin alone conferred a median survival of 20 days. Comparatively, carboplatin polymers offered 34 day median survival (p< 0.001) and dual agent polymer therapy corresponded with a 51 day median survival (p < 0.01 vs. carboplatin alone).
