Experimental hyperuricemia

Most species used for pharmacological experiments have rather low blood levels of uric acid. Experimental hyperuricemia can by induced by inhibition of the enzyme uricase. In most species uricase metabolizes uric acid to allantoin. Allantoxanamide blocks uricase and increases endogenously synthesized uric acid. This increase is blocked by compounds like allopurinol. [Pg.113]

Other mechanisms and mediators have been implicated in the genesis and prevention of chronic CSA nephrotoxicity. Reactive oxygen species, besides their functional effects on renal function, are also mediators of tissue injury favoring fibrosis. Use of the antioxidant vitamin E inhibited increases in TGF-beta and osteopontin mRNA and development of renal fibrosis in CSA-treated rats [184]. Lead-induced hyperuricemia may contribute to chronic lead nephropathy. Uric acid per se induces endothelial cell injury, renal microvascular disease, and hypertension, at least in part mediated by oxidative stress [38].
