Recently, there has been a growth of interest in the development of in vitro methods for measuring toxic effects of chemicals on the central nervous system. One approach has been to conduct electrophysiological measurements on slices of the hippocampus and other brain tissues (Noraberg 2004, Kohling et al. 2005). An example of this approach is the extracellular recording of evoked potentials from neocortical slices of rodents and humans (Kohling et al. 2005). This method, which employs a three-dimensional microelectrode array, can demonstrate a loss of evoked potential after treatment of brain tissue with the neurotoxin trimethyltin. Apart from the potential of in vitro methods such as this as biomarkers, there is considerable interest in the use of them as alternative methods in the risk assessment of chemicals, a point that will be returned to in Section 16.8. [Pg.305]

However, the effects were more marked for ginseng and were accompanied by reductions in frontal alpha waveband activity and decreased latency of the P300 component of the auditory evoked potential, a result that suggests more efficient stimulus assessment. [Pg.216]

Evoked potential studies showed that LSD facilitated a PSR this effect was blocked by cyproheptadine (18). The 5-HT agonist quipazine augmented a PSR this effect was blocked by cinanserin but (curiously) not by metergoline (80). Finally, tryptamine (183) and L-tryptophan (19) have been reported to increase PSRs in a cyproheptadine-reversible manner. [Pg.149]

Further support for the potentiating effects of Httexp on NMDAR activity was obtained by Lynn Raymond s and Michael Hayden s laboratories in the analysis of YAC72 HD mouse model (Hodgson et al., 1999). NMDA-evoked currents (Figure 4c, 4d) (Zeron et al., 2002) and NMDA-mediated Ca2+ transients (Zeron et al., 2004) were significantly increased in striatal neurons from YAC72 mouse when compared to wild type controls. Consistent with the HEK293 cells expression... [Pg.327]
