Aliphatic carboxylic adds are an important and widely used class of commercial chemicals. The commertial uses of carboxylic acids include synthetic intermediates, lubricants, catalysts, and preservatives, to name just a few. Some carboxylic adds cause liver (hepato) toxicity, most notably valproic add (2-propylpentanoic add) (12), and 2-ethylhexanoic add, (13). Although relatively few carboxylic adds cause liver toxicity, when it does occur it can be severe [49]. [Pg.87]

This has been found to be the case with valproic acid (2-propylpentanoic acid) (12), which is hepatotoxic, and 2-fluorovalproic add (22), which is much less hepatotoxic, discussed in Sedion 4.3.1. The hepatoxidty of 12 involves cytochrome P450 abstraction of its C-2 hydrogen atom. The C-2 fluorine atom of 22 cannot be removed by cytochrome P450 metabolism. It would be interesting to observe if the same isosteric replacement would reduce the hepatoxidty of other carboxylic acids, such as the widely used 2-ethylhexanoic add. [Pg.100]

Preparation 5-1 A mixture of 198 g of methyl methacrylate, 2 of methacrylic add, and 8 y of ferf-butylperoxy-2-ethylhexanoate is added drop-wise to 300 g of diethylene glycol diethyl ether kept at 100°C over a period of 1.5 h.
