M sodium hydroxide solution is added to the sample solution and the fluorescence is determined again. The addition of sodium hydroxide removes the fluorescence by ionising the phenol group of the ethinyloestradiol and thus any residual fluorescence which is due to excipients can be subtracted from the reading. In the BP assay the ethinyloestradiol content of the tablet extract is determined by comparison with the fluorescence of a solution containing a known amount of ethinyloestradiol standard analysed using the same conditions. 

Ethinyloestradiol and norethisterone, two synthetic hormones commonly used in oral contraceptives were analyzed in this study, both qualitatively and quantitatively. Tablets with a total mass of 80 mg, 0.05 mg ethinyloestradiol content, and 0.25 mg norethisterone content were used. Drug was extracted from the powdered samples with chloroform. Aliquots of the extracts were placed in an aluminum sample cup and scanned in triplicate with a Technicon InfraAlyzer 450 spectrometer.
