Eosin histological staining method

For morphological analyses, internal organs of mice were fixed in 10.0% neutral formalin they were processed by the standard histological technique and paraffin sections were stained in haematoxylin and eosin. Changes in internal organs of mice were assessed by a semi-quantitative method with divisions into four gradations (points) — not detected (0), l-t, 2+, 3-I-. 

Histology. The tissues listed in Table 21.6 plus any lesions, masses, or abnormal tissues are embedded, sectioned, and strained for light microscopy. Parafin embedding and staining with hemotoxylin and eosin are the preferred routine methods, but special stains may be used for particular tissues or for a more specific examination of certain lesions. Electron microscopy may also be used for more specific examination of lesions or cellular changes after their initial localization by more routine methods. 

The staining technique will depend on the type of cell of interest. Hematoxylin and eosin (H E) stain is the most widely used method in histology. Using this technique, basophilic white blood cells stain dark blue, eosinophilic white blood cells stain bright red, and neutrophils stain a neutral pink. 

Hemotoxylin-eosin staining protocol is used to observe histology of cancer tissue of treated and non-treated animals. Other parallel methods could be followed to reach the similar outcome. 

Histologic analysis of hematoxylin and eosin stained sections of liver and heart from male PPARa -/- mice following a single injeetion of etomoxir revealed marked lipid accumulation in both organs with high fatty acid oxidative flux (Fig. 3 A). The sections were also stained with oil red O, a method for staining of neutral lipids (Fig. 3B).
