As an example, chymotrypsin is classified EC 3.4.4.5 according to the IUB system.210 In this book a more mechanistically based classification is used. Because some official names are quite long, traditional trivial names for enzymes often have been retained. Remember that to be precise it is always necessary to mention the species from which an enzyme was isolated and, if possible, the strain. Also remember that almost every significant genetic difference is reflected in some change in some protein. It is possible that the enzyme you are working with is slightly different from the same enzyme prepared in a different laboratory. [Pg.498]

The hydroxynitrile lyase (HNL) class of enzymes, also referred to as oxynitrilases, consists of enzymes that catalyze the formation of chiral cyanohydrins by the stereospecific addition of hydrogen cyanide (HCN) to aldehydes and ketones (Scheme 19.36).275 279 These chiral cyanohydrins are versatile synthons, which can be further modified to prepare chiral a-hydroxy acids, a-hydroxy aldehydes and ketones, acyloins, vicinal diols, ethanolamines, and a- and P-amino acids, to name a few.280 Both (R)- and (.S )-selective HNLs have been isolated, usually from plant sources, where their natural substrates play a role in defense mechanisms of the plant through the release of HCN. In addition to there being HNLs with different stereo-preferences, two different classifications have been defined, based on whether the HNL contains a flavin adenine dinucleotide (FAD) co-factor. [Pg.383]

Forty four alkaloids were newly isolated from the 22 lupin plants and the structures were elucidated in our recent study. The structures were determined by spectroscopic investigations, in particular, by extensive 2D-NMR experiments in the recent works, and by chemical transformation. Biosynthetic pathways are also proposed, in some cases, by the preparation of isolated enzymes and by in vitro cell culture system. Clavulanic acid is a mold product with only weak intrinsic antibacterial activity, but it is an excellent irreversible inhibitor of most (3-lactamases. It is believed to acylate the aotive site serine by mimioking the normal substrate. Hydrolysis occurs with some (3-lactamases, but in many cases, subsequent reactions ooour that inhibit the enzyme irreversibly. This leads to its classification as a mechanism-based inhibitor (or so-oalled suicide substrate). The precise chemistry is not well understood (Fig. 38.18), but when clavulanio acid is added to ampicillin and amoxicillin preparations, the potenoy against (3-lactamase-produoing strains is markedly enhanced. [Pg.1605]

NSP, by definition, includes all the plant polysaccharides other than starch. For the reasons given above, NSP are divided into two broad classes in the classification and measurement scheme described here (Table 1) (1) the cell-wall NSP, which impart rigidity, and encapsulate and control the release of other nutrients, and (2) other NSP, including gums and refined preparations of cell-wall material, which occur in foods mainly as additives. There is no endogenous human enzyme for the hydrolysis of NSP, which are therefore nonglycemic, and all become available for fermentation in the large intestine. [Pg.468]
