Enzyme-lined immunosorbent assay

Marked peripheral eosinophilia is observed in almost all patients and nonspecific elevated serum immunoglobulin E (IgE) in about 75% of patients. ANCA can be detected in 38% to 50% of patients, yielding a perinuclear immunofluorescent label pattern (P-ANCA) with 75% to 81% of ANCA-positive sera. P-ANCAs are mostly directed against MPO as assessed by enzyme-lined immunosorbent assay (ELISA) (92-100% of the P-ANCA) (2,3). 

This chapter describes the procedures that can be used to determine compounds that have antiviral activity against HIV. These include maintenance of lymphoblastoid cell lines, preparation of peripheral blood mononuclear cells (PMMCs), and determination of the infectivity of the HIV stock-supernatant and antiviral assays. The assays described use both acutely and chronically infected cells. Toxicity of compounds is assessed by measuring 14C uptake. These protocols are used for the evaluation of compounds that can be carried out by a single individual in a Category 3 containment laboratory. The number of compounds analyzed would be about 10, which is a convenient number to fill a single 96-well p24 enzyme-linked immunosorbent assay (ELISA) plate. 

DNA-dependent protein kinase catalytic subunit, was obtained by cDNA cloning from a monoclonal antibody 18-2-ptoducing cell line 24). ScFvs were purified by conventional or anti-E tag affinity chromatognq>hy for use in enzyme-linked immunosorbent assay (ELISA), immunofluorescence assays, and microinjection experiments. 

Temperature sensors, such as the thermistors and thermopiles described in 4.4.4, can be used to monitor the variation in enthalpy arising from a reaction catalysed by an enzymatic label on either an andgen or an andbody. This technique is called TELISA (thermometric enzyme linked immunosorbent assay), and involves the use of reactor columns lined with antibodies, which detect antigens such as albumin [261]. These reactors, however, are not real biosensors.
