Electrophysiology dissection

Examine the cultures after 10-12 DIV to check for the preservation of pyramidal and granule cell layers. Discard cultures that display selective neuronal loss of a hippocampal region (see Note 15). Cultures may be used up to 28 DIV (see Note 16). Make electrophysiological recordings from each slice culture preparation to ensure that spontaneous activity does not anse prior to 28 DIV. Discard the entire dissection if spontaneous activity >0 5 mV in air litude is observed (see Fig. 2 and Note 17). [Pg.19]
