Electrical fields, generation electrophoresis

Electrophoresis due to the macroscopic electric field generated by the concentration gradient of the electrolyte whose anion and cation have unequal diffusion coefficients... [Pg.582]

Response to Electric and Acoustic Fields. If the stabilization of a suspension is primarily due to electrostatic repulsion, measurement of the zeta potential, can detect whether there is adequate electrostatic repulsion to overcome polarizabiUty attraction. A common guideline is that the dispersion should be stable if > 30 mV. In electrophoresis the appHed electric field is held constant and particle velocity is monitored using a microscope and video camera. In the electrosonic ampHtude technique the electric field is pulsed, and the sudden motion of the charged particles relative to their counterion atmospheres generates an acoustic pulse which can be related to the charge on the particles and the concentration of ions in solution (18). [Pg.549]

Generation of Heat in Electric Fields. One of the practical problems encountered in electrophoresis is the generation of heat from resistive dissipation of energy in the electrophoretic medium. The generation of heat (foule heating) is given by... [Pg.179]

The basic requirements of electrophoresis are a semi-conducting medium and an electric field. The semi-conducting medium can be a paper soaked in an electrolyte or a gel placed in an electrolyte. In the case of CE, it is a capillary filled with an electrolyte or a gel. The electric field is generated by applying a voltage difference over the capillary. [Pg.580]

Electrodriven separations, such as capillary electrophoresis (CE) and capillary electrochromatography (CEC), are based on the different electrophoretic mobilities in an electric field of the molecules to be separated. They provide a higher separation efficiency then conventional HPLC since the electrophoretic flow (EOF) has a plug-flow profile. Whereas the mobile phase in CE is driven only by the electro-osmotic flow, it is generated in CEC by a combination of EOF and pressure. CEC has a high sample capacity which favours its hyphenation with NMR. [Pg.242]

In the case of carrier electrophoresis, a sheet of paper, starch, polyacrylamide or agarose gel or similar materials, where the respective solid phase is saturated with buffer solution, are placed between the electrodes in order to generate a homogeneous, rectified electrical field. The mixture of substances to be separated is supplied as a single spot and the individual components are then separated in the course of time. Carrier electrophoresis is therefore a discontinuous separation process. [Pg.170]

Application of the electric field and the resultant resistance to current flow in the buffered paper generates heat. This is the greatest source of difficulty with paper electrophoresis. Heat dries the paper, which in turn leads to more resistance to current flow, which causes greater resistance, and so forth. [Pg.64]
