Since its conception, the dynamic filter has been widely reported and further developed. Most European designs are comprised of a multistage disk arrangement (Fig. 28) with both the rotating and stationary elements covered with filter cloth, thus utilising the space inside the pressure vessel. Such filters have been found (29) to be from 5 to 25 times more productive in mass of dry cake per unit area and time than filter presses for the same moisture content of the final slurry. In some cases, the moisture content with the dynamic filter was actually lower than with a filter press. Although not a taxane, ixabepilone is a novel microtubule inhibitor that was recently approved for metastatic breast cancer in combination with the oral fluoropyrimidine capecitabine or as monotherapy. It is a semisynthetic analog of epothilone B, and is active in the M phase of the cell cycle. This agent binds directly to 6-tubulin subunits on microtubules, leading to inhibition of normal microtubule dynamics. Of note, this agent continues to have activity in drug-resistant tumors that overexpress P-glycoprotein or tubulin mutations. Therefore, the pharmacokinetic parameters, which can be derived from blood level measurements, are important aids to the interpretation of data from toxicological dose-response studies. The plasma level profile for a drug or other foreign compound is therefore a composite picture of the disposition of the compound, being the result of various dynamic processes.
