Chemotherapeutic agents drugs

Several of the naturally occurring indoles also have clinical importance. The dimeric vinca alkaloid vincristine and closely related compounds were among the first of the anti-mitotic class of chemotherapeutic agents for cancer[14]. The mitomycins[15] and derivatives of ellipticine[16] are other examples of compounds having anti-tumour activity. Reserpine, while not now a major drug, was one of the first compounds to show beneficial effects in treatment of mental disorders[17]... 

There have been few reports on the cUnical use of albumin microspheres for the delivery of drugs. The limited clinical trials, using chemotherapeutic agents, employ a technique referred to as arterial chemoembolism in which the drug-containing microspheres... 

Intraperitoneal administration of chemotherapeutic agents has been used for many years as a way of increasing the delivery of drugs to tumors (e.g., ovarian carcinoma) located in the peritoneal cavity (Markman, 1986 Howell and Zimra, 1988). Cisplatin (Casper et al., 1983 Markman et al., 1985), cytosine arabinoside (Ara-C) (King et al., 1984 Markman et al., 1985, 1986), and bleomycin (Markman et al., 1986) are examples of intraperitoneally administered drugs which were already successfully applied in clinical settings.

Regrettably, all chemotherapeutic agents have the potential to produce adverse reactions with varying degrees of frequency and severity, and these include hypersensitivity reactions and toxic effects. These may be dose-related and predictable in a patient with a history of hypersensitivity or a previous toxic reaction to a drug or its chemical analogues. However, many adverse events are idiosyncratic and therefore unpredictable. 

One interesting example of the potential importance of intracellular pH gradients is found in the phenomenon of multidrug resistance (MDR). The MDR protein acts as a molecular pump, actively pumping chemotherapeutic agents out of the cell. Tumor cells, which are susceptible to chemotherapeutics have often a lower intracellular pH [178], which would lead to enhanced accumulation of these drugs in response to the pH differ-... 

Polyamines and their ammonium salts have been of interest because they are known to have potential applications as chelating agents (1-3), ion exchange resins (4-6), flocculants (7,8), and other industrial uses (9). Recent biomedical applications have constituted another important use of polymeric amines they have been investigated for use as biocompatable materials, polymeric drugs, immobilization of enzymes, cell-culture substratum and cancer chemotherapeutic agents (10-12).

A possible source for chemotherapeutic agents is the medicinal flora of the Asia-Pacific region. The purpose of this chapter is to provide a fundamental approach to understanding the potential of the medicinal flora of this region as a source of new anticancer drugs.
