Figure 16 (a) Structures of adenylation domain intermediates and inhibitors aminoacyl-sulfamoyl adenosine (AMS) and cisoid -like macrocyclic inhibitor, (b) Alkyne-functionalized chemical probe for NRPS A and PCP domains, (c) Structure of aminoacyl PCP, SNAC substrate analogue, and hydrolytically stable phosphopantetheinyl analogue, (d) Structure of vinylsulfonamide probe. R represents a peptide component and R an amino acid side chain. [Pg.649]

The catalytic subunit of cAPK contains two domains connected by a peptide linker. ATP binds in a deep cleft between the two domains. Presently, crystal structures showed cAPK in three different conformations, (1) in a closed conformation in the ternary complex with ATP or other tight-binding ligands and a peptide inhibitor PKI(5-24), (2) in an intermediate conformation in the binary complex with adenosine, and (3) in an open conformation in the binary complex of mammalian cAPK with PKI(5-24). Fig.l shows a superposition of the three protein kinase configurations to visualize the type of conformational movement. [Pg.68]

As special cases, both Eqs. (9.40) and (9.44) must be limits of a single function which also describes the intermediate situations in which the interior of the coil domain is partially penetrated by the pattern of flow. [Pg.611]

The crystal stmcture of the intermediate is not well understood. The final iron phase is termed superparamagnetic because the particle size is too small to support ferromagnetic domains. At low rates, the discharge occurs in two steps separated by a small voltage difference. At high rates, however, the two steps become one, indicating that the first step is rate limiting, ie, the second step (eq. 34) occurs immediately after formation of the intermediate (eq. 33). [Pg.535]

An Arrhenius plot of the rate constant, consisting of the three domains above, is schematically shown in fig. 45. Although the two-dimensional instanton at Tci < < for this particular model has not been calculated, having established the behavior of fc(r) at 7 > Tci and 7 <7 2, one is able to suggest a small apparent activation energy (shown by the dashed line) in this intermediate region. This consideration can be extended to more complex PES having a number of equivalent transition states, such as those of porphyrines. [Pg.108]

Figure 14.5 The domain organization of intermediate filament protein monomers. Most intermediate filament proteins share a similar rod domain that is usually about 310 amino acids long and forms an extended a helix. The amino-terminal and carboxy-terminal domains are non-a-helical and vary greatly in size and sequence in different intermediate filaments. In the ease of intermediate topography, they assume that there is a eertain eharaeteristie (for a given surfaee) length, r or, whieh defines the size of small domains eharaeterized by the same value of adsorption energy, and write the pair density distribution as... [Pg.254]

The first domain of one subunit of the fatty acid synthase interacts with the second and third domains of the other subunit that is, the subunits are arranged in a head-to-tail fashion (Figure 25.9). The first step in the fatty acid synthase reaction is the formation of an acetyl-O-enzyme intermediate between the acetyl group of an acetyl-CoA and an active-site serine of the acetyl trails-... [Pg.811]

If the vibration analyzer permits acquisition of time-domain data, additional time-waveform data should be obtained from the intermediate guide as well as the inlet and discharge valves. The intermediate guide is located... [Pg.722]

Another critical stage in the synthesis has been reached. The previous sections have summarized the reactions leading to the synthesis of both domains of calicheamicin y[, each with the correct absolute stereochemistry and in a form suitable for further advance. We are now in a position to describe the union of key intermediates 8 and 9 (see Scheme 21). [Pg.556]

Scheme 10.8 Biosynthesis of epothilone. Individual PKS domains are represented as circles and individual NRPS domains as hexagons. Acyl carrier proteins (ACPs) and thiola-tion domains (T) are posttranslationally modified by a phos-phopantetheinyl group to which the biosynthetic intermediates are covalently bound throughout the chain assembly. The thioesterase domain (TE) cyclizes the fully assembled carbon chain to give the 16-membered lactone. Following dehydration of Cl 2—Cl 3 to give epothilones C and D, the final step in epothilone biosynthesis is the epoxidation of the C12=C13 double bond by the cytochrome P450 enzyme P450epol<.
