Disorder propensity

The matrix Denv = [< env ((,/)] for the 119 kinases assayed (Fig. 9.2c) is obtained after inference of the dehydrons for the 87 kinases unreported in PDB from direct structure prediction. The predictions are validated through a correlation with an independent and accurate sequence-based prediction of the disorder propensity. This attribute was chosen because loopy regions, in the twilight between order and disorder (Chap. 5), compromise somewhat the accuracy of a structure prediction.

Concerns about psychotropic side effects, which can vary cross-culturally, often lead to premature cessation of psychotropics. This may be related to different propensities for and values placed on somatic experiences in different cultures. Transcultural research indicates that patients from non-Western cultures are more likely to present with predominantly somatic symptoms of psychiatric disorders (Ng, 1997 Parker, Gladstone 8c Chee, 2001), as cultural explanatory models and social demands may serve to bias information processing in the various domains of subjective experience (Angel 8c Thoits, 1987). All the disorder-specific depletions except one are, from the leftmost, typically buried amino acids. The one exception, N, is out of place in being both a surface-preferring and an order-promoting residue. Perhaps the short side chain, with its propensity to hydrogen-bond to the backbone (Presta and Rose, 1988 Richardson and Richardson, 1988), tends to induce local structure. This order-inducing tendency might explain the out-of-place behavior of N. 

Because of their propensity for being disordered, polymeric solids are some-times called organic glasses, but some polymers are inorganic, so this is not a good practice. 

In any case, through the propensity and specificity for aggregation (e.g., by jS-sheet formation in amyloids), proteins, silk, or otherwise, they will precipitate and form disordered aggregates or fibrils under the appropriate conditions (whether in vivo or in vitro Dobson, 1992 Fandrich et al., 2001 Uversky and Fink, 2004). 

OCD is also frequently comorbid with AN in fact, some researchers posit that the eating disorders are a modern variant of OCD peculiar to Western culture. In particular, the propensity for excessive exercise among some patients with AN may indicate the presence of comorbid OCD. Using a different set of statistical procedures, Kravitz et al. (2005) found that prescribing an antidepressant drug was 2.92 times more likely when a standardized patient presented with major depressive rather than adjustment disorder symptoms, and 8.50 and 10.3 times more likely when the patient made a brand-specihc or a general medication request, relative to no specific medication request, respectively. Physicians varied systematically in their propensity to prescribe an antidepressant, regardless of the type of patient presenting. But none of the standardized patients was systematically more or less likely than other patients to receive an antidepressant drug prescription. 

During the early twentieth century the barbiturates were used in children and adolescents for their sedative and hypnotic effects however, their safety profile and propensity to cause physical dependence led scientists in search of safer anxiolytics. The development of animal models of behavioral disorders facilitated the formulation of drugs with more specific central nervous system (CNS) effects. In 1959, chlordiazepoxide (Librium) was the first benzodiazepine (BZ) to receive a patent. It entered the market in 1960, followed by diazepam (Valium) in 1963. Today, over 35 BZs have been formulated and over 10 are available in the United States (Ballenger, 1995 Hobbs et ah, 1996). 

Because of their better side effect profile compared with clomipramine, including their lower propensity to decrease the seizure threshold, selective SRIs (SSRIs) have been receiving increasing attention as a treatment for the interfering symptoms associated with autistic disorder and other PDDs.
