In April 1988, the new kidney Dutch test (NKDT) replaced the Micrococcus (formerly Sarcina ) luteus kidney test. The NKDT is an one-plate test based on examination of the urine present in the renal pelvis by means of paper discs inserted in the renal pelvis (104). The test organism is Bacillus subtilis BGA and incubation is performed for 13-18 h at 37 C. The kidney is incised, and four paper discs are inserted and left there for 30 min. Two paper disks are placed diagonally on the surface of the test plate, and three control disks containing 0.5 g oxytetracycline, 0.05 g sulfamethazine, and 0.5 g tylosin are also placed in the middle of the plate. A previously reviewed method was applied for OTC, TC, CTC, DXC, and DMC analysis in tissue and egg samples however, further optimization and improvement were necessary. The optimal recoveries from tissue were obtained using succinate buffer and MeOH as a depro-teinization agent. The eluate from the MCAC column was acidified and further purified on an Empore disk equipped with a poly(styrene-divinylbenzene)-RP sulphonated membrane previously activated with MeOH and hydrochloric acid (pH 1.0). The elution of TCs was done with methanolic ammonia solution. The extract was evaporated under vacuum and reconstituted in oxalic acid solution. Even though the stability of TCs is poor under alkaline conditions, no influence on the recovery was observed (59-76% with RSD < 6.5% for kidney samples) (26).

Acaricidal and insecticidal activities of the synthesized compounds were determined as follows. In the case of two-spotted spider mite (T. urticae), female adult mites were placed on the excised leaf of kidney bean seedling, and then, the test solution (test confounds were dissolved in acetone at 20 to 200 ppm) was sprayed onto die leaf. Mortality was determined 2 days after the treatment. In the case of western flower thrips (F. occidentalis), first instar larvae were released on released on the excised leaf of kidney bean seedling (15 larvae per a disk), which was then sprayed with the test solutioa Mortality was determined 2 days after the treatment. In die case of silver leaf Miitefly B. argentifolii), adult whiteflies were released on die cabbage leaf and allowed to oviposit for 24 hours. Adults were removed from the leaf and the leaf was stood for 7 days. A leaf disk of 20 millimeters in diameter was excised from the leaf and sprayed with the test solution. Mortality was determined 6 days after the treatment. The acaricidal and insecticidal activities were indicated as activity indexes 3,2 and 1 which represent the mortality of 100%, 99% to 50% and less dian 49%, respectively.
