Notable examples of general synthetic procedures in Volume 47 include the synthesis of aromatic aldehydes (from dichloro-methyl methyl ether), aliphatic aldehydes (from alkyl halides and trimethylamine oxide and by oxidation of alcohols using dimethyl sulfoxide, dicyclohexylcarbodiimide, and pyridinum trifluoro-acetate the latter method is particularly useful since the conditions are so mild), carbethoxycycloalkanones (from sodium hydride, diethyl carbonate, and the cycloalkanone), m-dialkylbenzenes (from the />-isomer by isomerization with hydrogen fluoride and boron trifluoride), and the deamination of amines (by conversion to the nitrosoamide and thermolysis to the ester). Other general methods are represented by the synthesis of 1 J-difluoroolefins (from sodium chlorodifluoroacetate, triphenyl phosphine, and an aldehyde or ketone), the nitration of aromatic rings (with ni-tronium tetrafluoroborate), the reductive methylation of aromatic nitro compounds (with formaldehyde and hydrogen), the synthesis of dialkyl ketones (from carboxylic acids and iron powder), and the preparation of 1-substituted cyclopropanols (from the condensation of a 1,3-dichloro-2-propanol derivative and ethyl-... 

Useful syntheses of D- and L-lyxose from 1,3-O-benzylidene-D- and L-arabinitol have been achieved through the highly selective oxidation of the primary hydroxyl groups by dimethyl sulfoxide-dicyclohexylcarbodiimide.463 Oxidation of but one of the two (equivalent) hydroxyl groups in 1,3,4,6-tetra-O-benzyl-D-mannitol464 and l,6-di-0-benzyl-2,5-0-methylene-D-mannitol465 was possible with dimethyl sulfoxide-acetic anhydride. 

Primary alkyl halides (chlorides, bromides, and iodides) can be oxidized to aldehydes easily and in good yields with dimethyl sulfoxide.311 Tosyl esters of primary alcohols can be similarly converted to aldehydes,312 and epoxides313 give a-hydroxy ketones or aldehydes.314 The reaction with tosyl esters is an indirect way of oxidizing primary alcohols to aldehydes (9-3). This type of oxidation can also be carried out without isolation of an intermediate ester The alcohol is treated with dimethyl sulfoxide, dicyclohexylcarbodiimide (DCC),315 and anhydrous phosphoric acid.316 In this way a primary alcohol can be converted to the aldehyde with no carboxylic acid being produced. 

Oxidations using dimethyl sulfoxide activated by various reagents began with discoveries by Komblum and co-workers that primary tosylates and certain a-bromo ketones could be converted into aldehydes and glycoxals, respectively, by treatment with dimethyl sulfoxide as the oxidising agent. This was followed by the discovery several years later by Pfitzner and Moffatt that alcohols could be oxidised to carbonyl compounds with dimethyl sulfoxide, dicyclohexylcarbodiimide (DCC) and phosphoric acid at room temperature.2 Eventually the method developed by Swern and co-workers, involving activation of dimethyl sulfoxide with oxalyl chloride, came to be the most synthetically useful and widely applied of these mild oxidation procedures.3-6...

Aromatic a-diketones can be prepared from the acyloin compounds however, aliphatic diketones cannot be prepared by this method. The reaction proceeds well in complex systems without epimerization of adjacent stereocenters, as in the yohimbine example (eq 15). This method compares favorably with that of dimethyl sulfoxide-dicyclohexylcarbodiimide. 

Androst-4-ene-3,17-dione. Testosterone (0.58 g, 2 mmoles) is dissolved in a solution prepared from 3 ml of benzene, 3 ml of dimethyl sulfoxide, 0.16 ml (2 mmoles) of pyridine and 0.08 ml (1 mmole) of trifluoroacetic acid. After addition of 1.24 g (6 mmoles) of dicyclohexylcarbodiimide, the sealed reaction flask is kept overnight at room temperature. Ether (50 ml) is added followed by a solution of 0.54 g (6 mmoles) of oxalic acid in 5 ml of methanol. After gas evolution has ceased ( 30 min) 50 ml of water is added and the insoluble dicyclohexylurea is removed by filtration. The organic phase is then extracted twice with 5 % sodium bicarbonate and once with water, dried over sodium sulfate and evaporated to a crystalline residue (0.80 g) which still contains a little dicyclohexylurea. Direct crystallization from 5 ml of ethanol gives androst-4-ene-3,17-dione (0.53 g, 92%) in two crops, mp 169-170°.

The synthesis of L-ribofuranose derivatives from 16a has been carried out by Walker and Hogenkamp (35). The procedure involves oxidation of the hydroxymethyl group of 16a with dimethyl sulfoxide N,N dicyclohexylcarbodiimide and acid hydrolysis of the protecting group to give L-riburonic acid, which was converted into methyl (methyl a,/ -ribofurano-sid)uronates (26). Reduction of 26 with sodium bis(2-methoxyethoxy)alu-minum hydride gave the chromatographically separable anomers of methyl L-ribofuranoside (27). 

There are several methods reported in the literature for transforming vicinal diols into ct-diketones while avoiding the risk of C-C bond cleavage.26 Examples include the standard Swem conditions (dimethyl sulfoxide and oxalyl chloride followed by triethylamine), or the use of DMSO activated by acetic anhydride, pyridine-sulfur trioxide complex, or dicyclohexylcarbodiimide (Mq/J-att oxidation). Diones are also obtained by treatment with benzalacetone as a hydride acceptor in the presence of catalytic amounts of tris(triphenylphosphine)ruthenium dichlonde [(PPh RuCFl.27 Recent developments include the use of w-iodoxyben/.oic acid28 or the oxoammonium salt of 4-acctamidoletramethylpipcridine-1-oxyl and y -toluencNulfonic acid.29... 

Baker and Buss425 used the Pfitzner-Moffatt procedure (phosphoric acid-A /V -dicyclohexylcarbodiimide dimethyl sulfoxide) for the preparation of methyl 3-benzamido-4,6-0-benzylidene-3-deoxy-Q -D-uraZ) o-hexopyranosid-2-ulose and methyl 2-benzamido-4,6-(3-benzylidene-2-deoxy-a-i>n/ >o-hexo-pyranosid-3-ulose in yields of 90%.

The starting compound 251 was reduced to 252 with sodium borohydride. The latter was heated under reflux in 6% sulfuric acid in methanol to afford compound 253. Treatment of the latter with maleic anhydride at 170° for 3 hr afforded compound 254. Bisdecarboxylation of 254 with dicarbonyl bistriphenylphosphinenickel in anhydrous diglyme under nitrogen at reflux temperature for 6 hr afforded the olefin 255 in 69% yield (171). The latter was reduced with lithium aluminium hydride to the primary alcohol 256, which was oxidized to the aldehyde 257 with Ar,A -dicyclohexylcarbodiimide, dimethyl sulfoxide and pyridine in dry benzene. Treatment of the aldehyde 257 with an excess of the Grignard reagent prepared from l-bromo-3-benzyloxybutane afforded a mixture of diastereoisomers represented by the structure 258.

Diamino-3-carboxy-5-chloropyrazine in dimethyl sulfoxide (a) with N,N dicyclohexylcarbodiimide gave 2,6-diamino-3-chloro-5-(2, 5 -dioxopyrrolidin-l ylcarbonyOpyrazine ( ) (962), (6) with dicyclohexylcarbodiimide (and 2-hydrazino pyrimidine) gave 2,6-diamino-3-chloro-5-(A -cyclohexyl-V-cyclohexylcarbamoyl) carbamoylpyrazine (1331), and (c) as its triethylamine salt with V-ethyl-5-phenyl... 

Dimethyl sulfoxide (DMSO), which is successfully used to dehydrogenate primary alcohols to aldehydes, converts secondary alcohols into ketones in very high yields and under very gentle conditions. The mechanism is discussed in a previous section. Dehydrogenation and Oxidation of Primary Alcohols to Aldehydes (equation 217). The first oxidations were carried out in the presence of dicyclohexylcarbodiimide and an acid catalyst such as pyridinium trifluoroacetate [1016], which protonates the diimide and facilitates the attack by dimethyl sulfoxide (equation 259). 

In more recent works, the use of dicyclohexylcarbodiimide has been abandoned because the reaction works satisfactorily with acid catalysts alone, followed by bases such as triethylamine or diisopropylethylamine, which gives, sometimes, even better yields than triethylamine [1012,1023. Several activators are being used, and the best seems to be oxalyl chloride (theSwern oxidation) [1023,1149]. Other activators are mentioned in the section Oxidation of Primary Alcohols to Aldehydes. The advantages of oxidations with dimethyl sulfoxide lie in the mildness of the reagent and in the low temperatures, sometimes -45 °C [1020] or -60 °C [1023], at which the reactions are run. 

To a refluxing solution of 4 g (0.033 mol) of 4,6-octadiyn-l-ol and 21 g (0.10 mol) of dicyclohexylcarbodiimide in 200 mL of absolute ether, a solution of 1.7 g of crystalline phosphoric add in 100 mL (110 g, 1.41 mol, 43 x molar excess) of dimethyl sulfoxide is added dropwise. After an additional heating for 4 h, 100 mL of 4 N sulfuric acid is added. The precipitated crystals of A/,A/ -dicyclohexylurea are filtered with suction and washed with ether. The filtrate is washed until neutral with a solution of sodium hydrogen carbonate. The dried ether solution is evaporated, and the residue is distilled at 60 °C at 0.001 mm of Hg to give a 73% yield of 4,6-octadiynal as a colorless oil. 

Selective oxidation of amylose at C3 has been achieved with dimethyl sulfoxide/phosphorus pentaoxide [119]. In contrast, the oxidation of unprotected cellulose with dimethyl sulfox-ide/acetic anhydride [120] or dicyclohexylcarbodiimide/dimethyl sulfoxide [121] produced a product containing modified 8-D-glucopyranosyl units - some with a carbonyl function at C2, some with a carbonyl function at C3, and some with carbonyl functions at both C2 and C3[122],...
