A mixture of 4.98 g of acetoacetic acid N-benzyl-N-methylaminoethyl ester, 2.3 g of aminocrotonic acid methyl ester, and 3 g of m-nitrobenzaldehyde was stirred for 6 hours at 100°C in an oil bath. The reaction mixture was subjected to a silica gel column chromatography (diameter 4 cm and height 25 cm) and then eluted with a 20 1 mixture of chloroform and acetone. The effluent containing the subject product was concentrated and checked by thin layer chromatography. The powdery product thus obtained was dissolved in acetone and after adjusting the solution with an ethanol solution saturated with hydrogen chloride to pH 1 -2, the solution was concentrated to provide 2 g of 2,6-dimethyl-4-(3 -nitrophenyl)-1,4-dihydropyridlne-3,5-dicarboxylic acid 3-methylester-5- -(N-benzyl-N-methylamino)ethyl ester hydrochloride. The product thus obtained was then crystallized from an acetone mixture, melting point 136°Cto 140°C (decomposed).

Condensation of aromatic methylesters such as methyl 4-methoxybenzoate 351 a or methyl 4-hydroxybenzoate 351b with excess sodium-HMDS 486 in a mixture of THF-l,3-dimethyl-imidazolin-2-one (DMEU) at 185 °C in a closed vessel affords 59 or 93% of 4-hydroxybenzonitrile 298 as well as 26% 352 with smooth cleavage of the aromatic methyl ether in 351a (Scheme 4.47). Methyl indole-3-carboxylate gives hkewise 3-cyanoindole in 81% yield [127] (cf. also ref [92] in section 4.3).

Auch mit Alkylphosphaten ist eine 1,5-regioselektive Alkylierung moglich. So werden z.B. 1,2-Dimethyl-5-nitro- (63%) und l-Ethyl-2-methyl-5-nitro-imidazol (74%) durch Umsetzung von 2-Methyl-4(5)-nitro-imidazol bei 160 ° mit Polyphosphorsaure-methylester bzw. -ethylester erhalten868. 

In einer ahnlichen 1,3-dipolaren Cycloaddition erhalt man aus 1,5-Dimethyl-pyrazinium-3-olat und Acrylsaure-methylester 6-Methoxycarbonyl-8-methyl-4-methylen-2-oxo-3,8-diaza-bicyclo[3.2.1 ]octan3. 

Das aus l-tert.-Butyl-2-methoxycarbonyl-l-trimethylsiloxy-cyclopropan mit Titan(IV)-chlorid in situ gebildete Keten-Derivat reagiert mit N,N-Dimethyl-methaniminium-chlorid in Gegenwart von katalytischen Mengen Trifluormethansulfonsaure-trimethylsilylester unter Bildung von 5,5-Dimethyl-2-dimethylaminomethyl-4-oxo-hexansaure-methylester (89%) ... 

Durch Umsetzung von 1,3,5-trisubstituierten Hexahydro-l,3,5-triazinen mit 1-Organoxy-1-trimethylsilyloxy-l-alkenen in Gegenwart von Trifluoressigsaure erhalt man in zumeist guten Ausbeuten 3-Amino-propansaure-ester, z.B. 2,2-Dimethyl-3-isopropylammo-propansaure-methylester (83%) und 3-Allylamino-2,2-dimethyl-propansaure-methylester...
