Dihydroergotamine

Lisuride dihydrogen maleate (h/Jj 30-35), methysergide maleate (h/ij 40-45), dihydroergotamine mesylate (hRf 45-50) and ergotamine tartrate (hRj 70-75) appeared as blue violet chromatogram zones on a yellow background. The detection limits — calculated for free base — were 3-4 ng substance per chromatogram zone.

Dihydroergotamine (DHE) is available for intranasal and parenteral (IM, IV, subcutaneous [SC]) administration. Patients can be trained to self-administer DHE by the IM or SC routes. 

A solution of dihydroergotamine (254) methanesulphonate in water was left in the sunlight for 15 days. The only photolysis product found was the derivative (255). There had been no alcohol in the reaction solution, so the molecule was assumed to have formed an oxo-derivative which gave the enol ether during separation by thick-layer chromatography [158], (Scheme 2.6). 

Over the years it has been interesting to note that many compounded products eventually become commercially available products. Recent examples might include fentanyl lozenges, minoxidil topical solution, nystatin lozenges, clindamycin topical solution, tetracaine-adrenalin-cocaine (TAG) solution, dihydroergotamine mesylate nasal spray, buprenorphine nasal spray, buffered hypertonic saline solution, and erythromycin topical solution as well as numerous other dermatological and pediatric oral liquids and some...
