Digoxin detection

Bioluminescence can also be used as the basis for immunoassay. For example, bacterial luciferase has been used in a co-immobilized system to detect and quantify progesterone using a competitive immunoassay format (34), and other luciferase-based immunoassays have been used to quantify insulin, digoxin, biotin, and other clinically important analytes (35).

Detection and result The dried chromatogram was immersed in the dipping solution for 2 s and heated to 110°C for 10 min. Observation in long-wavelength UV light revealed fluorescent zones for digoxin (h/ f 30-35) blue and digitoxin (h/ f 40-45) yellow. 

Homogeneous electrochemical enzyme immunoassays for both phenytoin and digoxin have been developed. In both cases the label was glucose-6-phosphate dehydrogenase, which catalyzes the reduction of NAD to NADH. The NADH produced was detected by LCEC at a carbon paste electrode.

Including in the medication a biological marker that is non-toxic, inert, chemically stable and easily detectable in biological fluids (such markers include riboflavin, phenol red and small quantities of digoxin). 

Tablets-A single initial dose of 500 to 750 meg (0.5 to 0.75 mg) of digoxin tablets usually produces a detectable effect in 0.5 to 2 hours that becomes maximal in 2 to 6 hours. Additional doses of 125 to 375 meg (0.125 to 0.375 mg) may be given cautiously at 6- to 8-hour intervals until clinical evidence of an adequate effect is noted. The usual amount of digoxin tablets that a 70 kg patient requires to achieve 8 to 12 mcg/kg peak body stores is 750 to 1250...

The treatment of an overdosage of digoxin requires monitoring of cardiac rhythm in order to detect arrhythmias. 

Determining the optimal level of digitalis effect may be difficult. Unfortunately, toxic effects may occur before the therapeutic end point is detected. Measurement of plasma digoxin levels is useful in patients who appear unusually resistant or sensitive a level of 1 ng/mL or less is appropriate.
