Although EPO deficiency is the primary cause of CKD anemia, iron deficiency is often present, and it is essential to assess and monitor the CKD patient s iron status (NKF-K/DOQI guidelines). Iron stores in patients with CKD should be maintained so that transferrin saturation (TSAT) is greater than 20% and serum ferritin is greater than 100 ng/mL (100 mcg/L or 225 pmol/L). If iron stores are not maintained appropriately, epoetin or darbepoetin will not be effective, and most CKD patients will require iron supplementation. Oral iron therapy can be used, but it is often ineffective, particularly in CKD patients on dialysis. Therefore, intravenous iron therapy is used extensively in these patients. Details of the pharmacology, pharmacokinetics, adverse effects, interactions, dose, and administration of erythropoietin and iron products have been discussed previously. [Pg.985]

Fresh and frozen human tissue samples obtained from brain, liver, and kidney have been analyzed for hydrogen sulfide levels by sulfide-derived methylene blue determination using ion-interaction reversed-phase HPLC (Mitchell et al. 1993). This method can quantify nmol/g levels of sulfide. Gas dialysis/ion chromatography with ECD has been utilized for measurement of sulfide in brain tissue with 95-100% recovery (Goodwin et al. 1989). [Pg.158]

Neurotransmitters, peptides, and other endogenous substances in the extracellular space can be sampled using in vivo microdialysis. The dialysis membrane typically excludes the transport of larger molecules and enzymes that could otherwise interfere with analysis of the substances of interest. In the case of neurotransmitters, the levels in dialysates are the net result of the interaction between processes affecting release into and removal from the extracellular space. Consequently, in vivo microdialysis can only sample a neurotransmitter that has not yet been removed by clearing mechanisms and this method does not provide information regarding intracellular levels of substances. [Pg.223]

The equilibrium dialysis experiment revealed that histidine-substituted salicylamide was selected as an RNA ligand. Subsequent binding analysis by UV titrations and Job plot revealed the histidine-substituted salicylamide Cu + complex bound the target RNA hairpin with an apparent dissociation constant of 150 nM. This binding constant likely reflects more complex binding processes than a simple 1 1 interaction, as the observed binding curve saturates well below the concentration of the histidine-substituted salicylamide, and thus the actual affinity of the complex for targeted RNA is probably lower.
