Detergent CHAPS

Tools have recently been developed that should enable identification of the catalytic subunits of the plasma membrane / -(l,3)-glucan synthase from red beet storage tissue and other plant sources. These include an efficient procedure for solubilization and enrichment of this enzyme using the detergent CHAPS, generation of antibodies, and use of affinity labels such as UDP-pyridoxal. 

Many protein samples require the use of detergents for their solubilization. For IEF work, the zwitterionic detergents CHAPS and CHAPSO, or the nonionic detergent octyl-glucoside, at concentrations of 1-2% in the gel are recommended. Even in the presence of detergents, some samples may have stringent salt requirements and may aggregate in salt-free environments.

The desaturase (and following cyclase) enzymes are integral membrane proteins that can be solubilized by the detergent CHAPS. Enzymatic activity is lost when the enzymes are solubilized, but it is restored when reconstituted in proteoliposomes (Britton, 1993). In the first phase of desaturation, the conversion of (15Z)-phytoene into (15Z)- -carotene occurred in the dark, with oxygen as an essential requirement. The reaction was independent of cofactors such as NADP and NADPH. (15Z)- -Carotene could not be desaturated further, but when this compound was isomerized by light, the isomeric mixture formed underwent further desaturation to produce lycopene, not...
