The convergent approach has seen extensive use in the construction of functional macromolecules because of its ability to modify dendrons at both the focal point and the chain ends. This modularity is especially helpful in design optimization because the same dendritic structure can be modified, after the dendron synthesis, to vary the number and chemistry of functional moieties in the resultant dendrimer.72...

Ethers are chemically and coordinatively more inert than amides. Indeed, Frechet-type polyether dendrimers prepared in solution demonstrate a broad spectrum of chemical and coordinative compatibilities. Accordingly, polyether dendron synthesis on solid support was developed, initially by Bradley and coworkers and later by Portnoy and Dahan Instead of the 3,5-dihydroxy benzyl alcohol derivatives used in the soluble Frechet-type dendrimers, a more SPOS suitable 3,5-bis(hydroxymethyl)phenol unit was conceived as a building block for the dendron construction.

HPLC is limited to relatively low molar mass compounds, Le. below 3000. In practice this tends to restrict the technique to dendrons rather than fully formed dendrimers, but it is nonetheless a useful technique, given the importance of establishing high analytical purity of products at each step in a dendrimer synthesis.

A dendrimer-based approach for the design of globular protein mimics using glutamic (Glu) and aspartic (Asp) acids as building blocks has been developed [151]. All the fullerene-containing dendrimers reported to date have been prepared with a Cgo core but never with fullerene units at their surface or with Cgg spheres in the dendritic branches. We have recently started a research program on the synthesis of dendrons substituted with fullerene moieties. These fulleroden-drons are interesting building blocks for the preparation of monodisperse fullerene-rich macromolecules.
