For the synthesis of perfectly dendronized sohd-phase polymers (Fig. 7.4) various dendritic structures were prepared based on amide connections [6]. For example, the naturally occurring amino acid lysine was used as a building block in creating a dendritic scaffold [33]. The synthesis of symmetrical tri-branching den-drimers on aminomethyl polystyrene macrobeads was also described in literature [34]. The existence of a hollow core as well as a densely packed exterior layer in PAMAM dendrimers was proven by studying their conformational behavior [3]. Meijer et al. introduced the concept of dendritic boxes by synthesis and characterization of dendrimer with flexible core and solid shell structure. The flexible core was based on poly(propyleneimine) dendrimer. The rigid shell was obtained by modification of terminal groups with bulky amino acid derivatives, that is (r-Boc)-protected L-phenylalanine. Dendrimer structure was fully studied by H and CNMR, IR and UV techniques. Additionally, solid-phase behavior of the shell was confirmed by spin-lattice (Ti) and spin-spin (T2) relaxation measurements and molecular mechanics calculations [3,18,19]. 

In this paper we will focus more specifically on the third one, in which the scaffold unit is originated from monosaccharides. Compare to classical Pamam, Boltorn cores, carbohydrate units add much more 3D information coded in the chirality in almost each carbon. The diversity of such stmcture has been calculated for oligosaccharides and is veiy impressive, Laine found over 1.05 x 10 possible branched hexasaccharides starting from a basis set of 6 different sugars. Compared with a set of 6 different amino acids, there is nearly 10 times less hexapeptides Thereby the possible structures of a carbohydrate-based dendrimers are tremendous and open a large possible choice of scaffolds and thus properties. In addition, various characteristics of such dendrimer can be easily controlled such as their sizes, shapes, topologies, flexibilities and their surface properties.
