All three zones can convert cholesterol into pregnenolone, but because of the presence/absence of certain enzymes, the flow of pregnenolone is preferentially directed to the synthesis of the steroid for which the particular zone is specific. It has not been determined whether the zona fasciculata produces dehydroepiandrosterone (DHEA) and dehydroepiandrosterone sulphate (DHEAS) in physiologically significant amounts.

C]taurocholic acid (201) and [7-3H]dehydroepiandrosterone sulphate (202) in [2H5]glycerol matrix have been used184 to interpret the partial negative ion FAB spectra of these unlabelled compounds and to show that FAB and field desorption mass spetrometry enable one to determine quickly the specific radioactivity and label distribution within highly labelled biochemicals. The minimal specific radioactivities detectable by this method are about 20MBq/mmol and lOGBq/mmol for compounds labelled with 14C and with tritium, respectively, with 1% and 5% accuracy. The use of mass spectrometry minimizes interferences from labelled impurities.

In 23 healthy aged subjects (73.0 8.0 years), the impaired hypothalamo-pituitary-adrenal function became apparent in a most significant decline of the plasma levels of dehydroepiandrosterone and dehydroepiandrosterone-sulphate (Martinez-Ta-BOADA et al. 2002). A close correlation between immune changes with ageing and dehydroepiandrosterone response to ACTH stimulation was found. There was an inverse correlation of lymphocyte changes with the plasma levels of steroids, especially dehydroepiandrosterone and its metabolite, dehydroepiandrosterone-sulphate.

Testosterone and oestradiol are known as the sex steroid hormones. Testosterone is the principal androgen and is synthesized by the testes in the male. Oestradiol. which is secreted by the ovaries, varies w idely in concentration in plasma throughout the female menstrual cycle. Steroids with oestradiol-like action arecalledoestrogens. Progesterone is a product ofthe ovary and is secreted when a corpus lutcum forms after ovulation. Normal female plasma also contains testosterone, about half of which comes from the ovary and half from peripheral conversion of androstenedione and dehydroepiandrosterone (DHA) sulphate which are secreted by the adrenal cortex. Some oestradiol is present in low concentration in normal male plasma. 

An effective antiserum for dehydroepiandrosterone 3-sulphate has been obtained by use of the 17-hydrazone as hapten, coupled to BSA. 3a,5p Tetrahydroaldosterone (22), the main urinary metabolite of aldosterone, is efficiently bound by an antiserum induced by the conjugate of BSA with the 3-carboxymethyloxime of aldosterone 18,21-diacetate. Haptens for RIA of cortisone and cortisol have been prepared by the introduction of 7a- and 7/3-carboxymethyl substituents (23) through a malonic ester condensation with the 7-bromo-derivative (24). ...
