Culture media composition comparison

The medium for N. mediterranei seed culture were glucose, 20 g/L yeast extract, 5 g/L bactopeptone, 5 g/L malt extract, 5 g/L (pH 7.3 adjusted with 1 N NaOH). The seed culture was carried out as in the EL coli culture except the temperature (30°C). The WAC of the BC-chitosan composite from biosynthesis was enhanced approximately 120% in comparison with that of the BC film. The improvement of the WAC was affected by increasing the MW and concentration of chitosan in the culture medium [73]. 

For comparison purposes, Table 5.1 shows the composition of DMEM (one of the most versatile medium formulations, typically employed in the culture of mammalian cells) and Schneider s medium (used for diptera insect cells, especially Drosophila melanogaster). A total of 34 different components are present in DMEM, although some minor variations in composition may occur between suppliers. 

Regarding the components of bulk fermentation processes, the strain of the organism used to manufacture the drug substance for the clinical study should be compared with the strain to be used in commercial production. Strain identification includes microbiological, cultural, and biochemical characteristics. A comparison of the media composition and method of sterilization, sterilization parameters, and the pH of the medium after sterilization should be done. All fermentation stages, parameters, and conditions should be described in detail (i.e., temperature, pH) and documented.
