All static studies at pressures beyond 25 GPa are done with diamond-anvil cells conceived independently by Jamieson [32] and by Weir etal [33]. In these variants of Bridgman s design, the anvils are single-crystal gem-quality diamonds, the hardest known material, truncated with small flat faces (culets) usually less than 0.5 nun in diameter. An impressive example of the application of structure-based methods was the design of a inhibitor of the HIV protease by a group of scientists at DuPont Merck [Lam et al. 1994 This enzyme is crucial to the replication of the HIV virus, and inhibitors have bee shown to have therapeutic value as components of anti-AIDS treatment regimes. The star1 ing point for their work was a series of X-ray crystal structures of the enzyme with number of inhibitors boimd. Their objective was to discover potent, novel leads whid were orally available. Solubility Properties. Fats and oils are characterized by virtually complete lack of miscibility with water. However, they are miscible in all proportions with many nonpolar organic solvents. Tme solubiHty depends on the thermal properties of the solute and solvent and the relative attractive forces between like and unlike molecules. Ideal solubiHties can be calculated from thermal properties. Most real solutions of fats and oils in organic solvents show positive deviation from ideaHty, particularly at higher concentrations. Sintering has been used to produce a porous polytetrafluoroethylene (16). Cellulose sponges are the most familiar cellular polymers produced by the leaching process (123).
