Costunolides

The intramolecular Hiyama reaction is excellently suited for the construction of carbocyclic and heterocyclic ring systems of different size. In most examples, problems of induced diastereoselectivity are involved (Sections 1.3.3.3.9.2.2. and D.2.3.). In the total synthesis of the antitumor germacranolide ( )-costunolide from ( ,.E)-farnesol, the correctly substituted ( , )-cyclodecadiene moiety was constructed by an intramolecular Hiyama reaction22. 

Vanillosmopsis erythropappa Schultz-Bip. is a component of the Atlantic coastal forest in Brazil, where it is exploited as a source of an essential oil used in the pharmaceutical industry. A survey of specimens from four locations showed markedly different compositions of their essential oil fraction (Lopes et al., 1991). The compounds identified were bisabolol [160], costunolide [161], eremanthine [162], and five related compounds, six compounds based upon the cyclocostunolide skeleton [163], and the two esters 15-deoxygoyazenolide [164] and lychnopholide [165]... 

From the fruits extracts of L. nobilis, costunolide, santamarm, reynosin, gazaniolide, 11,13-dehydrosantonin and spirafolide were isolated as known sesqniter-pene lactones (Fig. 6.7). The dichloromethane fraction also afforded a new sesquiterpene, named lauroxepine, which had oxepine type sesqniterpene stractnie as an derivative of spirafolide [20]. However, its activity was the least (IC = 34.6... 

There is little information on the stability of the sesquiterpene lactones. However, one report [21] states that a chloroform solution of parthenolide stored at room temperature was found to contain costunolide diepoxide (29) after a few days, probably formed by aerial oxidation. The literature also suggests that parthenolide can polymerize on prolonged storage [22].

Brominative cyclization. Treatment of either costunolide (1) or deydro-saussurca lactone (2) with NBS in aqueous acetone at 2(T results in the cyclized hromo lactones 3. 4, and 5, in approximately the same ratio from both substrates.3... 

The family Lauraceae consists of 50 genera and 2000 species of trees, shrubs, and herbs, of which 70 are of medicinal value in the Asia-Pacific region. Lauraceae are well-known for elaborating isoquinoline alkaloids and sesquiterpenes, the latter most likely representing a vast source of material for the search for NOS. Examples of such compounds are costunolide and dehydrocostunolide found in the leaves of Laurus nobilis (bay leaf, laurel), the leaves of which are widely used as a spice, antiseptic, stomachic, and to treat rheumatism in traditional European medicine (58). The potential of Neolitsea zeylanica Nees (Merr.) as a potential source of NOS inhibitor is discussed here.
