It is often observed that when PCR products from ancient remains are cloned, they contain a large number of random substitutions which are believed to be due to damage present in extracted DNA.10 When this error frequency is compared to that of a control template of modem, presumably undamaged DNA, the increased number of errors itself may indicate that the PCR products stem from a damaged template and thus that it likely represents an old sequence.13 However, it should be remembered that if the presumed ancient sequence in reality stems from a contaminating modem sequence present in only a few copies, the PCR product will have gone through many more cycles of polymerization before the plateau phase of the amplification is reached. For example, if an amplification product contains twice as many errors as a control DNA amplified for 30 cycles, that result could be due to errors in the initial template or to the presence of template in a 10- to 100-fold lower concentration (A. von Haeseler and S. Paabo, unpublished, 1991). Thus, because the concentration of ancient DNA is hard or impossible to determine accurately, error rates in amplification products may be difficult to use as a criterion of authenticity. [Pg.418]

Filtration Filtration (qv) is appHed in blood cell separation to remove leukocytes from ted blood cell (RBC) and platelet concentrates. Centtifugational blood cell separators do not reduce white blood cells (WBC) in red cell and platelet products sufficiently to avoid clinical complications such as GvHD and alloimmunization. A post-apheresis filtration step is needed to further reduce the WBC load. Modem filters are capable of a 3-log reduction in white cell contamination of the blood product, eg, apheresis single-donor platelet units having a typical white cell contamination of 5 x 10 white cells in 4 x 10 platelets can be reduced to a 5 x 10 white cell contamination, a sufficiently low number to avoid severe transfusion reactions. [Pg.523]

Modem real time instmmental methods permit analyses of unstable transient species and the free-radical intermediates as well. These methods have gready expanded the scope and power of VPO studies, but important basic questions remain unresolved. Another complication is the role of surface. Peroxide decompositions and radical termination reactions can occur on a surface so that, depending on circumstances, surfaces can have either an inhibiting or accelerating effect. Each surface has varying amounts of adventitious contaminants and also accumulates deposits during reaction. Thus no two surfaces are exactly alike and each changes with time. [Pg.338]

Another thermally efficient kiln is the modem mixed-feed vertical kiln ia which coke is admixed with 8.5—20 cm lump limestone and charged into the top of the vertical kiln by a mobile, overhead charging system. When a sample of ca 100 g has been obtained, a representative sample for use in size characterization equipment must then be taken. Some of the more modem methods of size characterization require as Htde as 1 mg of powder, thus obtaining a representative sample can be quite difficult. If the powder flows weU and does not contain too many fines, a device known as the spinning riffler (Fig. 4c) can be used. A spinning riffler consists of a series of cups that rotate under the powder supply. The time of one rotation divided into the time of total powder flow should be as large a number as possible. Although this device has been shown to be very efficient, problems can be encountered on very small (1 mg) samples, and the powder must be processed several times. Moreover, in order to avoid cross-contamination, cleanup after each of the sampling processes can be quite difficult. The goal of filtration in the modem municipal treatment plant is a maximum of 0.1 ntu (nephelometric turbidity unit), which ensures a sparkling, clear water (8). Crop yields can rise dramatically with the use of commercial fertilizers. For example, in 1800 an acre of land in the United States produced about 25 bushels of com. In the 1980s the same acre of land produced 110 bushels. Worldwide, approximately 4 billion acres of land are used to grow food crops. This would probably be enough land to feed the world s population if the entire acreage could be fertilized commercially. It has been estimated that world crop production would increase by about 50% if about 40 per acre were spent to apply modem chemical fertilizers.
