Clofibrate Clofibrate, ethyl ether 2-(4-chloropheoxy)-M( -butyric acid (20.2.2), is synthesized by esterifying 2-(4-chlorophenoxy)-/yo-butyric acid (20.2.1) with ethyl alcohol. This is synthesized in a single-stage reaction from 4-chlorophenol, acetone, and chloroform in the presence of an alkali, evidently by initial formation of chlorethone-trichloro-tert-butyl alcohol, which under the reaction conditions is converted into (4-chlorophenoxy)trichloro-fert-butyl ether, and further hydrolyzed to the desired acid 20.2.1, which is further esterified with ethanol in the presence of inorganic acid [6-8]. [Pg.272]

Activated alumina (260 °C) column has been used for isolation of PCDEs, PCDDs, and PCDFs from chlorophenol extracts [37,38]. Four fractions were collected from an alumina column (50 g) petroleum ether (400 ml), 5% ethyl ether in petroleum ether (200 ml), 25% ethyl ether in petroleum ether (400 ml), and ethyl ether (400 ml). To prepare the [m-(acetylpyridinio)-n-alkyl]adenosine pyrophosphates, 10 mmoles of acetylpyridinio-n-alkylphosphoric acid and 10 mmoles of adenosine 5 -phosphoromorpholidate (as salt of 4-morpholine iV,iV -dicyclohexylcarboxamidine) are dissolved in 20 ml of freshly distilled o-chlorophenol. The mixture is stored at room temperature for 7 days. Progress of the condensation reaction is checked daily by paper electrophoresis (0.1 M Tris chloride at pH 8.1, 30 V/cm). After the reaction is completed, 60 ml of water are added to the mixture and the suspension is extracted 3 times, each with 200 ml of ethyl ether. The first ether extract obtained is reextracted with 30 ml of water. The combined aqueous phases are reduced to a volume of 5 ml under reduced pressure and are charged on to a Dowex 1X8 column (formate form, 2 X 50 cm, 100-200 mesh). Typical procedure A mixture of p-chlorophenol (128 mg, 1 mmol), NaOH (40 mg, 1 mmol) and THF (10 mL) was stirred for 10 min allyldiisobutylteUuronium bromide (360 mg, 1 mmol) was then added. The reaction mixture was stirred for another 5 h at room temperature under nitrogen. Aqueous saturated NaHCOj solution was added and extracted with CH2CI2. The extract was dried over anhydrous MgS04 and concentrated in vacuo. The residue was chromatographed on sihca gel with 95 5 hexane-ethyl acetate as eluent to give a colourless oil of allyl p-chlorophenyl ether (145 mg, 86%). [Pg.225]
