Centrifugal partition chromatography

Actual droplet formation is not necessary as long as the mobile phase can pass through the stationary phase without displacing it. As a matter of fact, both aqueous and nonaqueous biphasic systems have been used (51, 332). Recent results reveal that a variety of natural products such as saponins, saturated fatty acids, and biomacromolecules - the latter on a biphasic aqueous polymer system - can be separated (332). Because of its comparatively rapid and non-destructive action, CPC has recently been used in the isolation of the tunichromes, the rather sensitive phenolic peptides from the vanadium-collecting sea squirt Ascidia nigra (53, 54) and of W-cis- and trans-rciindA from a complex photoisomerization mixture (51). [Pg.45]

Centrifugal partition chromatography (CPC) has been used to characterize the partitioning behavior of hydrophilic molecules, where log D values as low as —3 can be obtained [371,377-379]. It is not as popular a method as it used to be, apparently due to instrumental challenges. Cyclic voltammetry (CV) has become the new method used to get access to very low log D values, with partition coefficients reported as low as —9.8 [261,269,362]. [Pg.55]

A number of techniques have been used for the preparative separation of flavonoids. These include HPLC, Diaion, Amberlite XAD-2 and XAD-7, and Fractogel TSK/Toyopearl HW-40 resins, gel filtration on Sephadex, and centrifugal partition chromatography (CPC). The choice of methods and strategies varies from research group to research group and depends often on the class of flavonoid studied. [Pg.2]

Centrifugal partition chromatography (CPC) relies on centrifugal force rather than gravitation for the retention of the stationary phase and solvents can be pumped at higher speeds through the instruments. In addition, no need for droplets formation is required. This allows shorter separation times, without loss of resolution, and an infinite choice of solvents with the only requirement of forming two immiscible phases, stable with the time. Chloroform-based systems have been mostly used for the separation of saponins due to their favourable partition coefficient towards such solvents. [116, 119, 120]. [Pg.207]

Centrifugal Partition Chromatography (CPC) has been shown to yield partition coefficients which compare quite favorably with those measured by traditional shake-flask (El Tayar, 1993). The procedure, which employs a coil planet type centrifuge in a horizontal flow-through system, has been described (El Tayar, 1991) The equipment can handle organic solvents with low viscosity (such as heptane) more readily than it handles octanol, but log octanol/water values in the range of -3.0 to +3.0 seem reliable. It has been used quite effectively in measuring the log Ps of amino adds (Tsai, 1991). [Pg.110]
