If a solution forms part of an electrochemical cell, the potential of the cell, the current flowing through it and its resistance are all determined by the chemical composition of the solution. Quantitative and qualitative information can thus be obtained by measuring one or more of these electrical properties under controlled conditions. Direct measurements can be made in which sample solutions are compared with standards alternatively, the changes in an electrical property during the course of a titration can be followed to enable the equivalence point to be detected. Before considering the individual electrochemical techniques, some fundamental aspects of electrochemistry will be summarized in this section. 

In this paper the fundamental aspects of process development for the production of core and virus-like particles with baculovirus infected insect cells are reviewed. The issues addressed include particle formation and monomer composition, chemical and physical conditions for optimal cell growth, baculovirus replication and product expression, multiplicity of infection strategy, and scale-up of the process. Study of the differences in the metabolic requirements of infected and non-infected cells is necessary for high cell density processes. In the bioreactor, the specific oxygen uptake rate (OURsp) plays a central role in process scale-up, leading to the specification of the bioreactor operational parameters. Shear stress can also be an important variable for bioreactor operation due to its influence on cell growth and product expression. 

In this paper the fundamental aspects concerning the production of CLPs and VLPs with baculovirus infected insect cells are reviewed. It is not the goal of this communication to review all the aspects of insect cell culture technology, since this would be a task impossible to achieve in a single chapter. A fundamental aspect of biochemical organizations is that the enzymes that catalyze a sequence are often clustered together in the cell. As a result, an intermediate produced in the first reaction in a pathway is passed directly to the second enzyme in the pathway, and so on. Such an arrangement might be expected to accelerate synthesis of the end product, minimize the loss of intermediates, and facilitate regulation of the pathway.
