CD8 mAb

Immunomagnetic beads Dynabeads M-450 coated with sheep antimouse IgG, sheep or rat antimouse IgG subclass (Fc-specific), or preconjugated with MAbs specific for CD4 and CD8 (available from Dynal, Wirral, Merseyside, UK) A broad range of other preconjugated beads are now supplied by Dynal and are summarized m Note 2. They will remain stable for up to 1 yr when stored at 2-8°C Thiomersal (0 01% [w/v]), or sodium azide (0.02% [w/v]) is used as preservative. Hazard warning Both sodium azide and thiomersal are irritants to the skm and eyes, and are toxic if inhaled or ingested. Handle with care. 

An alternative approach for some human cell-surface markers (CD4, CD8, CD 19, and CD34) is the use of Detachabead, which is an anti-Fab polyclonal antiserum that causes dissociation of bound MAbs preconjugated to Dynabeads (see Note 9). For protocols detailing use of Detachabead see refs. 23—33. 

Detachabead is available from Dynal for positive selection of CD4+, CD8+, CD 19+, and CD34+ cell subsets (23-30) This reagent has also been used to select positively for CD45+ cells (31), HLA-DR+ Langerhans cells (32), and c-kit+ human lung mast cells (33) However, its suitability for dissociation of beads coated with MAbs of other specificities should always be assessed by the user in a pilot study. 

From these data it might be expected that removal of CD8 T cells would enhance IgE production and favour TH2-type immune responses. However, this appears to depend on the stage at which depletion is carried out. Thus, if CD8 T cells are depleted in rats (using the mAb OX-8) in which an IgE response had been induced, then the usual decline in serum IgE levels is prevented (Holmes etal., 1994). This supports the view that CD8 T cells suppress IgE. However, if depletion of CD8 T cells is performed prior to immunization, then the IgE response is not enhanced.
