Knapp et al.21 reported that the levels of forebrain tryptophan, serotonin (5-HT), 5-hydroxyindoleacetic acid (5-HIAA), and hydroxylase cofactor (BH4) were comparable in two experimental mouse strains (A/J and C57B1/6J), despite two- to three-fold differences in vitro in the relative activities of forebrain and midbrain tryptophan-5-monoxygenase. The enzyme activities did not differ with respect to Km for cofactor at saturating levels, but manifested different degrees of cooperativity with respect to cofactor when examined with BH4 concentrations within a physiological range. [Pg.23]

Strain differences in the activity of tryptophan hydroxylase, the rate-limiting enzyme in serotonin biosynthesis, have been reported in rats22-23 and in mice.24 Chaouloff et al.22 reported that the activity of tryptophan hydroxylase was decreased in Lewis rats compared with Fisher 344 rats. Kulikov et al.23 reported that in vitro central tryptophan hydroxylase activity was higher in the Lewis strain of rats than in spontaneously hypertensive rats. In mice, Knapp and Mandell24 reported that midbrain tryptophan hydroxylase from two behaviorally different mouse strains, C57B1/6J and A/J, had different stabilities and regulatory properties. [Pg.23]

With respect to mice, the CD-I is by far the most commonly used strain in the pharmaceutical industry. Other strains used less frequently are the B6C3F1, CF-1, NMRI, C57B1, Balb/c, and Swiss (PMA, 1988 Rao et al., 1988). Swiss is the generic term since most currently used inbred and outbred strains were originally derived from the Swiss mouse. [Pg.300]

Different strains of mice exhibit different susceptibilities to TCDD (dioxin) and other polycyclic hydrocarbons. Thus the C57B1 6 mouse is more responsive to substances such as TCCD and 3-methylcholanthrene, which interact with the AhR receptor and induce enzymes such as CYP1A1, whereas the DBA/2 mouse is less responsive. This has been found to be inherited as an autosomal dominant trait. The nonresponsive allele Ahd expresses a protein with diminished affinity for binding ligands such as TCDD. [Pg.146]
