Bovine plasma fibrinogen

Materials. Bovine plasma albumin (Lot. 12F-9365) and bovine plasma y-globulin (Lot. 116C-0147), 99 %electrophoretically pure, were purchased from Sigma Chemical Co., and bovine plasma fibrinogen (Lot. 26), 95% clottable, was purchased from Miles Laboratory Inc. The concentration of these plasma proteins was determined by spe-ctrophotometiy, and their absorption coefficients were taken to be as follows =6.67 for albumin (a) =11.2 for globulin (S) ... 

Components/ mechanism of action Human plasma, fibrinogen and thrombin, virally inactivated, hemostat, sealant. Autologous fibrinogen -t-platelet-rich plasma, hemostatic gel. Bovine collagen, bovine thrombin, plus autologous human plasma, hemostatic agent. Bovine collagen and bovine thrombin. Expands 20% which aids in hemostatic effect. 

The adsorption method of fibrinogen is as follows each of the different PU beads was first contacted with bovine plasma containing C-labeled human fibrinogen as a function of adsorption time or plasma concentration. Sample was then rinsed with phosphate buffered saline (PBS, pH 7.4, 0.15M) twice and placed in 2% sodium dodecyl sulfate (SDS) solution for 2 days, and the surface concentration of fibrinogen was determined by counting the radioactivity. 

The fibrinogen content of bovine Fraction I is generally 75% or better, refleeting the much higher concentration of fibrinogen in bovine plasma (52, 104). 

Protein samples used to conduct a protein transfer were bovine serum albumin (BSA, 67,000 daltons), chicken egg ovalbumin (Ovalb, 45,000 daltons), and human serum cryo-precipitate. These agents were obtained from plasma containing a mixture of proteins including Fibrinogen (340,000 daltons), human serum albumin (HSA, 67,000 daltons), and immunoglobulin G (IgG, 47,000-56,000 daltons). The cryo-precipitate was diluted with 20 ml of buffer solution prior to separation. 

In the study, we used bovine fibrinogen obtained from purified citrated blood plasma [13], Fibrinogen was transferred to 0.06 M phosphate buffer (pH 7.4) containing 0.15 MNaCl using Sephadex G-25 gel filtration.
