Because they are weak acids or bases, the iadicators may affect the pH of the sample, especially ia the case of a poorly buffered solution. Variations in the ionic strength or solvent composition, or both, also can produce large uncertainties in pH measurements, presumably caused by changes in the equihbria of the indicator species. Specific chemical reactions also may occur between solutes in the sample and the indicator species to produce appreciable pH errors. At the EM level, detection usually involves using a probe (oligonucleotide) in which a hapten has been incorporated. Incorporation of the hapten does not interfere with the hybridization of the complementary sequences. The next step is the binding of a reporter (may be an antibody) to the hapten. The reporter is then subjected to a binding molecule (may be a secondary antibody) that is coupled with an electron-dense material such as colloidal gold for visualization. The important forces involved in the adsorption of metals on to particles are attractive electrostatic or van der Waals forces. These concepts explain many of the properties of colloids with respect to the adsorption of contaminants or ion-exchange factors and the aggregation of the colloids into larger particles. These larger particulates may then descend the water column to the sediment. This occurs most notably in estuarine environments, as increases in salinity lead to estuarine silting. Binding of electrolytes to hydrophobic colloids is often used to facilitate their coagulation and precipitation. [Pg.362]

Using a glassy carbon electrode modified with a mercury film, Weber et al. [66] measured the association and dissociation rate constants for the complex formed between Pb + and the 18-crown-6 ether. It was found that Pb + forms a complex with 18-crown-6 with a stoichiometiy of 1 1 in both nitrate and perchlorate media. The formation constant, for the nitrate and perchlorate systems are (3.82 0.89) X 10 and (5.92 1.97) x lO mol Ls , respectively. The dissociation rate constants, are (2.83 0.66) x 10 with nitrate and (2.64 0.88) x 10 s with perchlorate as counter ion. In addition, the binding of Pb + with benzo-18-crown-6 embedded in a polymerized ciystalline colloidal array hydrogel has been also analyzed [67]. [Pg.45]
