Potassium benzyl penicillin

Crystalline antibiotic, moderately hygroscopic. Dec 214-2 IT. [ojff +285-310 (c = 0.7). Freely sol in water, in isotonic sodium chloride soln, and in dextrose solns. Sol in methanol, ethanol, glycerol. pH of 3% aq soln 5.0 to 7.5. Solutions stored at refrigerator temps remain stable for several days. They are rapidly inactivated by acids, alkali hydroxides, and by oxidizing agents. One milligram of benzyl penicillin potassium is equivalent to 1595 units. One international Or U.S.P. penicillin unit is equivalent to 0.6 micrograms of benzylpenicillin sodium.

A total of 650 ml of distillate is collected. The batch is allowed to cool to 50°C and then filtered. The crystals are washed with isobutyl acetate and then dried at 35°C for 24 hours. The virtually anhydrous penicillamine hydrochloride obtained weighs about 128 g (69% from potassium benzyl-penicillin).

Requires no refrigeration when dry. Aq solns may be kept for 3 days at 1 O 1 without significant loss of activity. Behrens prepn assayed 1630 units/mg. Less toxic than benzyl -penicillin in exptl animals.

In contrast to the sodium or potassium salt of benzyl penicillin, which is very water soluble and has a short duration of action, the procaine and benzathine salts (or more accurately, the ion-pair complexes) shown in Figure 8.27 are only sparingly soluble in water and very lipophilic. They are dissolved in an oily vehicle, such as ethyl oleate or sesame oil, and administered by intramuscular injection to create a depot from which the penicillin anion is slowly released into the bloodstream, achieving a sustained-release profile. An added advantage of these depot penicillins is their superior stability compared to benzylpenicillin. 

Other chemicals which inhibit milk lipase include hydrogen peroxide, animal cephalin, sodium arsenite, diisopropyl fluorophosphate, 2,4 din-itro-l-fluorobenzene, p-hydroxymercuribenzoate, potassium dichromate, lauryl dimethyl benzyl ammonium chloride, aureomycin, penicillin, streptomycin, and terramycin (Schwartz 1974).
