Bacterial screening

Coprinus micaceus is abrmdant in the prairie region of Canada and is an edible mushroom. The methanohc extract of C. micaceus exhibited anti-bacterial activity against A. faecalis, S. aureus, Corynebacterium xerosis and E. coli at the concentration of 250 pg/ml in a preliminary anti-bacterial screening assay. A chromatographic fractionation of methanohc extract of this mushroom subsequently resulted in the isolation of a new bioactive sterol, micaceol (12).

Valuable bacterial screening tests have been developed in the laboratories of Ames (41) and Rosenkranz (42). The Ames test measures back mutation to histidine independence of histidine mutants of Salmonella typhymurium and can be conducted with strains that are also repair deficient, that possess abnormalities in the c ell wall to make them permeable to carcinogens, or that carry an R factor enhancing mutagenesis. Phenol Coefficient Test. The first important attempt at standardizing testing methods was known as the phenol coefficient test (96). It has been modified several times, and is an official AO AC screening test recognized by EPA and PDA. The phenol coefficient test compares the activity of disinfectants to that of phenol, under specific conditions, to give a number that measures the activity of the chemical tested with respect to that of phenol, ie, the phenol coefficient. The AO AC method employs visual examination of bacterial growth in a nutrient medium. The Kelsey-Sykes test (1969) is a modified method popular in British circles.

The aim of our investigation was the development of the amperometric enzyme immunosensor for the determination of Klebsiella pneumoniae bacterial antigen (Ag), causes the different inflammatory diseases. The biosensing pail of the sensors consisted of the enzyme (cholinesterase) and antibodies (Ab) immobilized on the working surface of the screen-printed electrode. Bovine seiaim albumin was used as a matrix component. 

For example, a screening of 416 strains (71 bacterial strains, 45 actinomycetes, 59 yeast, 60 basidiomycetes, 33 marine fungi, and 148 filamentous fungi) has been performed to look for microorganisms that display reductase activity in the absence of oxidase activity [8b]. A new microorganism, Diplogdasinospora grovesii IMI... 

Extensive screening methodology was used to find the suitable microorganism. As a result, K. pneumoniae IFO 3319 out of 450 bacterial strains was found to give the corresponding (2J, 3S)-hydroxy ester with 99% de and >99% ee in kilogram scale quantitatively [29g].
