Bacillus megatherium

The simplest pyrimidine antibiotic is bacimethrin, 5-hydroxymethyl-2-methoxypyrimidin-4-amine (985), which was isolated in 1961 from Bacillus megatherium and is active against several yeasts and bacteria in vitro as well as against staphylococcal infections in vivo it has some anticarcinoma activity in mice (69MI21301). It may be synthesized by LAH reduction of ethyl 4-amino-2-methoxypyrimidine-5-carboxylate (984) which may be made by primary synthesis in poor yield, or better, from the sulfone (983) (B-68MI21304). 

By fermentation with Streptomyces griseus, S. olivaceus, S. aureofaciens. Bacillus megatherium or Propionobacterium freudenreichii. Molasses is used generally as fermentation medium, C0CI2 and 5,6-dimethylbenzimidazole are added. Various adsorption and extraction methods are used for isolation from the fermentation liquors. 

The basic microbial inhibition assay format involves a standard culture of a test organism, usually Bacillus stearothermophilus, Bacillus subtilis, Bacillus cereus, Micrococcus luteus, Escherichia coli, Bacillus megatherium, or Strepto-793... 

The CAST uses Bacillus megatherium and a 16-24 h incubation at 44-45 C, whereas STOP employs Bacillus subtilis and a 16-24 h incubation at 27-29 C. A zone of inhibition around the swab suggests the presence of a microbial inhibitor in the sample. In the US, bob veal calf carcasses are condemned on the basis of a positive CAST without further confirmation. For FAST, the organism and temperature are the same as those for the CAST, but the CAST medium is supplemented with dextrose and bromocresol purple. The faster growth rate of bacteria with FAST, allows reduction of minimum incubation from 16 to 6h. 

Abou Aly and Gomaa (2002) evaluated the effect of inoculation with diazotrophs (Azotobacter chroococcum or Azospirillum brasilense) combined with either Bacillus megatherium var. phosphaticum or Glomus mosseae, in the presence of half the recommended dose of N, on the growth and yield of coriander. Bacimethrin (5-hydroxymethyl-2-methoxypyrimidin-4-amine) was isolated from Bacillus megatherium. Its antibacterial activity spectrum covers several forms of yeasts and bacteria in vitro, and in vivo bacimethrin is active in staphylococcal infections. Anticarcinoma activity in mice has been demonstrated <69Mi 602-oi>.

Baelmathrin, 4-amino-2-me koxy-S-pyrimUUne-methanol an antibiotic synthesized by Bacillus megatherium. It is antagonized by thiamin and pyridoxine, and is active against some yeasts and bacteria.
