Assembly reactions

The reaction block, which provides a fully enclosed reaction environment (Teflon, polypropylene, and silicone rubber seals) is a slightly modified version of a design first disclosed by Sphynx Pharmaceuticals [23]. Figure 9 shows the reaction block and the reagent vials (100, 30, and 10 mL sizes) in their fully assembled and disassembled states. Floles at the bottom of the wells of the 96-deepwell polypropylene plate (sealed in fully assembled reaction block) allow the reaction solutions to be removed from the wells (via a separate vacuum plenum) and the functionalized resin (retained by Teflon frits) to be washed with solvents. [Pg.45]

In addition to the previously noted heterocycle syntheses under ring assembly, reaction of a cationic P-bromo-1 -aryl-2,3-dihydro-1.3.2-di i/ iphoxphininiiim salt 28 (X=Br) with sodium hydroxide in toluene gave the corresponding P-hydroxy derivative (see Sect. 4.3) [89],... [Pg.96]

Acyl-allylboronates are smoothly provided through a palladium-mediated three-component assembly reaction of an acyl chloride, an allene and a diboron with high regioselectivity and E-stereoselectivity (Scheme 16.56) [62]. [Pg.948]

Finally, one should recognize that determinations of the critical concentration depend wholly on the validity of the equilibrium or steady-state assumptions. If a stable end point for prdtomer-polymer coexistence is not attained, then kinetic factors affect the observed behavior. With the well recognized tendency of tubulin to lose its ability to engage in assembly reactions upon storage even at low temperature, and with the presence of various nucleotide hydrolases and transphosphorylases in microtubule protein, such kinetic effects are a serious problem. [Pg.190]

Fig. 7. (a) Example chromatin assembly curve recorded at 1.3 pN force applied to the magnetic bead. A magnification of the squared portion of the curve is also presented, (b) Assembly curve of the same DNA molecule, subjected to difference forces in the course of a single assembly round (forces were adjusted by adjusting the distances between the cuvette and the external magnet as indicated). The lines drawn indicate that the assembly rate changes as a function of force. Note the immediate response of the assembly reaction to the changing force. [Pg.386]

In contrast to the use of self-assembly reactions and metal ion coordination preferences to direct the construction of mixed cofactor systems, the use of SPPS or selective chemical ligation allows for the direct covalent attachment of cofactors for the construction of mixed cofactor systems within de novo design. Figure 11 shows the flavocy-tochrome maquette constructed by Dutton and co-workers (149) using a flavin moiety covalently attached to a unique cysteine residue inside a four helix bundle with bis-histidine binding sites for heme... [Pg.431]

Other Fe-S Clusters. Spontaneous self-assembly reactions using phosphines as terminal ligands have been reported to yield a variety of new Co-S (22) and Ni-S clusters (23). Extension to Fe-S clusters was initially effected by the following reaction in heterogeneous solution (2A). [Pg.267]

Synthetic efforts at preparing model complexes for the FeMo-cofactor have largely focussed on two types of Mo-Fe-S cluster, both of which are prepared via self-assembly reactions using tetrathiomolybdate as starting material. The first of these is the "linear" type of cluster, containing the MoS2Fe unit formed by coordination of discrete MoS - units to Fe. The second is the... [Pg.282]

Polynuclear Fe-M-S Complexes from "spontaneous self assembly" reactions. Synthetic analog clusters for the Fe2S2 and Fe4S4 centers in the Fe/S proteins (ferredoxins) have been obtained by procedures that are based on the concept of "spontaneous self assembly". The latter (30) assumes that the cores of the Fe/S centers are thermodynamically stable units that should be accessible fiom appropriate reagents even in the absence of a protein environment. [Pg.392]

Fleming and coworkers have developed a number of such multicomponent assembling reactions using alkyl halides, aldehydes, ketones and acid chlorides as electrophiles for post-functionalization of carbomagnesiation . The stereoselectivity of the reaction with electrophiles is not only generally high but also highly dependent on the nature of electrophiles. [Pg.671]

Place the assembled reaction flask in the water bath, attach the stirring motor, and begin stirring. [Pg.39]
