The artery was loaded with Fluo-4 as the membrane-permeant acetoxymethyl ester. Some of this high-affinity, Ca2+ indicator dye is often sequestered in the SR (cf. Goldman et al 1990). The SR can then be readily visualized, especially when [Ca2+]CYx is low (as in the panels at 0 and 6.8 s), because the intra-SR dye is saturated with Ca2+, and fluoresces brightly. This artery was treated with 1.0 fim phenylephrine (PE), which caused the [Ca2+]CYT level to oscillate asynchronously in the cells seen in the centre of the panel. The cell outlines are clearly visible when [Ca2+]CYT tiscs, as in the panels at 3.4 and 10.2 s. Note that nearly all of the SR (the very bright areas, especially in the 0 and 3.4 s panels) lies parallel to, and immediately beneath the PL (from Miriel at al 1999, with permission). [Pg.130]

A second distinguishing feature of /l-solenoids is their inter-coil interactions. Their stacked coils are coordinated by a network of H-bonds. Usually, the coils in a given /1-solenoid have similar cross-sectional shapes, an arrangement that allows corresponding strands in successive coils to form parallel /1-sheets. The term /1-arc has been proposed for the turns between strands (Hennetin et al., 2006) and they also stack with inter-coil H-bonds, yielding so-called /1-arcades. The inter-coil distance (4.8 0.2 A) is determined by the distance between H-bonded /1-strands, which is the same on all sides of the solenoid. Consequently, /1-solenoids are straight and they contrast in this respect with other kinds of solenoids which mostly have curved shapes (Groves and Barford, 1999 Kobe and Kajava, 2000). [Pg.59]

Strand-turn-strand motifs in /1-solenoids differ fundamentally from those found in globular proteins. In globular structures, two adjacent strands with an intervening /l-turn form an antiparallel structure called a /1-hairpin (Fig. 10A). In /1-solenoids, the polypeptide chain also folds back on itself, but the flanking /1-strands make contact via their side chains rather than interacting via H-bonds of the backbone (Fig. 10A). As a result, consecutive strands find themselves in two different, parallel, /1-sheets. The latter strand-turn-strand structure is called a /1-arch, and its turn, a /1-arc (Hennetin et at., 2006 Yoder and Jurnak, 1995). In /1-solenoids, /1-arches stack in-register to form /1-arcades which have two parallel /1-sheets assembled from corresponding strands in successive layers. [Pg.77]

The track ball is not as common a device as are the others in this section. Physically it is a ball that is mounted and can be rotated in any direction [the arcade version of Missile-Command uses this device]. As the ball rotates - the direction of rotation indicates the desired direction of movement - the position of a tracking device can be read by the computer (using 8 or 10 bit A/Ds) and a cursors position updated to give visual feedback.
