Columns antibody affinity

Many precolumns and trap cartridges for sample clean-up are commercially available. In our experience, a 2 to 3 cm short column with twice the analytical column inner diameter and packed with the same particles performs satisfactorily. An antibody affinity column for selective removal of highly abundant proteins from human serum samples provides better sensitivity for the discovery of low abundance protein markers that may represent revolutionary therapeutic diagnosis and monitoring. [Pg.372]

To investigate the effects of the different mass transfer mechanisms, breakthrough curves were generated on model monoclonal antibody affinity columns with two types of packings Sepharose 4B (Pharmacia) and controlled-pore glass (Electronucleonics, mean pore size 1273 ft) Mouse monoclonal anti-benzenearsonate IgG was produced in this laboratory by batch culture in a 15 L fermentor. The IgG was purified... [Pg.121]

Food Analysis Preparative immunoaffinlty chromatography techniques are also applicable to the analysis of food samples for the presence of parent aflatoxins. To validate this method, naturally contaminated corn and peanut product samples were obtained and extracts made using methanol-water (60% 40%, vol/vol). An aliquot of extract was applied to the monoclonal antibody affinity column, aflatoxins eluted, and the product measured by reversed phase HPLC. In Figure... [Pg.211]

SIAB and sulfo-SIAB have been used to make a high-capacity RNA affinity column for the purification of human IRP1 and IRP2 (Allerson et al., 2003), to couple antibodies or Fab fragments to amine-modified microparticles (Harma et al., 2000), and in the attachment of oligonucleotides to surfaces for detection arrays (Adessi et al., 2000). [Pg.289]

The Agilent multiple affinity removal system utilizes the specificity of antibody-antigen recognition for 14 highly abundant proteins from human serum samples. The affinity column achieves reproducible and specific depletion from human serum and plasma to eliminate 94% of interfering proteins. It allows identification of proteins down to nanograms per milliliter level as reported by Agilent. [Pg.372]

Fig. 4. Classification of reported noncompetitive immunoassays for haptens based on the assay principle. (A) Assays that include a chemical modification of hapten to allow sandwich-type detection. (B1) Improved single-antibody immunometric assays that separate immune complex and excess labeled antibody, either by using a hapten-immobilized affinity column or based on differences in their physical properties. (B2) A variation of single-antibody immunometric assays based on masking of unoccupied antibody by an immunoreactive macromolecule followed by selective capture and detection of the hapten-occupied antibody. (C) Assays employing a probe molecule specific to a hapten-antibody complex. Fig. 4. Classification of reported noncompetitive immunoassays for haptens based on the assay principle. The binding between antibody and antigen is generally quite tight thus, relatively harsh conditions are normally required to subsequently elute antigen off immuno-affinity columns. Several standard approaches may be adopted in this regard. Elution, using a buffer of low pH, is... [Pg.368]
