Anti-anxiety agent

The oldest anti-anxiety agent is undoubtedly alcohol and it is certain that this drug is still routinely self-administered for this purpose. Towards the end of the eighteenth century, bromide salts were used to relieve conditions akin to anxiety despite the risk of a characteristic toxic delirium, known as bromism . Alternative treatments, such as paraldehyde and chloral hydrate, were also widely used but these too had adverse effects the former can cause psychosis but the latter is still used as a sedative and anaesthetic agent.

To speed the process of drug discoveiy, combinatorial chemistry has been developed to prepare what are called combinatorial libraries, in which anyrwhere from a few dozen to several hundred thousand substances are prepared simultaneously. Among the early successes of combinatorial chemistry is the development of a benzodiazepine library, a class of aromatic compounds much used as anti-anxiety agents.

Anti-Anxiety Agents — Cartazolate (SQ 65, 396 11) at 100 mg daily was less effective than diazepam at 20 mg on various anxiety rating scales49 and GPA 2640 (12) at 1100—1300 mg daily failed to alleviate anxiety in anxious non-psychotics.50 Trazodone (13) at 75—150 mg daily was equivalent to 30—60 mg of chlordiazepoxide in relieving symptoms caused by anxiety.51...

Introduction During 1966, several new types of chemical structures were reported to have central nervous system depressant activity, and further studies were described of compounds previously classified in this area. The problem of defining further the type of activity to predict whether a given depressant will be of clinical application as an antipsychotic, anti-anxiety agent, sedative, hypnotic, anti-convulsant or anesthetic continues to occupy the attention of many investigators.
